| Objective:From the point of view that the similarity of mechanism and symptoms between TCM Blood Syndrome and cerebral malaria,we try to explore the effect of TCM method of cooling blood and dispersing blood on cerebral malaria,and to observe Xijiao(Shuiniujiao)Dihuang Tang and its separation’s effect on the cerebral malaria sequestration theory and inflammation theory,and to explore its possible role Mechanism.Methods:84 C57 BL / 6 female mice were randomly divided into six groups: blank group(Group z),model group(group M),artesunate group(Group q),Liangxue Sanxue group(Group x),Liangxue group(group L)and Sanxue group(Group s).The Liangxue Sanxue group was treated with Xijiao Dihuang Decoction,the Liangxue group was treated with Shengdi and Shuiniujiao in Xijiao Dihuang Decoction,and the Sanxue group was treated with Radix Paeoniae Rubra and cortex moutan in Xijiao Dihuang Decoction.From the second day after infection,artesunate was injected intraperitoneally at the concentration of 25 mg / kg in each group except the blank group and the model group.The last three groups were given 0.2ml by gavage once a day according to the groups On the 7th day of infection,9 mice in each group were selected.Blood was taken from eyeballs and brain tissues.Chemokines CXCL9 and CXCL10,inflammatory factors IFN-γ,IL-10 and IL-6 were detected,TNF-αwas used as the main observation index of blood cooling method,TSP-1,TSP-2 and CD36 were used as the main observation indexes of blood dispersing method,and the expression of ZO-1 occludin,ICAM-1 and VCAM-1 adhesion factors were observed.Pathological examination was performed in 3 mices.Result:1.The results of Rmcbs survival rate and infection rate Artesunate group,Liangxue Sanxue group and its separations could reduce the infection rate of ECM mice,prolong the survival time and improve the rmcbs score.The rmcbs score of ECM mice in Liangxue Sanxue group was higher than that of other groups on the 11 th day(P < 0.05)2.The results of q PCR:The gene expression level of ifn-y in brain tissue of model group was significantly higher than that of normal group,artesunate group,Liangxue Sanxue group and its its separations(P < 0.05).The gene expression level of ifn-y in brain tissue of Liangxue Sanxue group was significantly lower than that of artesunate group(P < 0.05).3.The results of WB:The expression level of tight junction protein ZO-1 in the normal group was significantly higher than that in the model group,artesunate group,Liangxue Sanxue group and its separations(P < 0.05),indicating that the other groups of tight junction protein ZO-1 were damaged in varying degrees.The protein expression level of ZO-1 in Liangxue Sanxue group was significantly higher than that in artesunate group and model group(P < 0.05).4.Theresults of ELISA :The expression levels of CXCL9 and CXCL10 in model group were significantly higher than those innormal group,artesunate group,Liangxue Sanxue group and their split groups(P < 0.05).There was no significant difference between artesunate group and Liangxue Sanxue group,and between the separations group and normal group.Conclusion1.Artesunate can inhibit the expression of chemokines CXCL9,CXCL10 and IFN-γ.2.Xijiao Dihuang Decoction Combined with artesunate can effectively inhibit the expression of IFN-γ.And Xijiao Dihuang Decoction Combined with artesunate has protective effect on the injury of tight junction protein ZO-1.3.According to the observation of this experiment,the curative effect of the split group is not obvious,and there is no special regulatory effect on the corresponding mechanism.Xijiao Dihuang Decoction,as the represent-ative prescription of the method of cooling blood and dispersing blood,as a whole of linkage and mutual cause,will weaken its effect after split. |
