Chemical Constituents From Plant Of Aspidopterys Obcordata Hemsl.

Aspidopterys obcordata Hemsl.is a plant of aspidopterys of Malpighiaceae,also known as"hei gai guan”.Its vines and stems are used as medicine,which is light,astringent and slightly cold.It has the effects of clearing fire,detoxifying,diverting water and removing stones,and is mainly distributed in my country’s Hainan,Yunnan and other places.Dai people are often used in the treatment of acute and chronic nephritis,postpartum weight loss,and endless lochia.At present,there are few reports about the chemical constituents of the A.obcordata Hemsl.In order to further understand the chemical constituents of the A.obcordata Hemsl.and find the active substances,the material basis of the plant was studied systematically.Mainly using silica gel column chromatography,ODS-C₁₈reversed-phase column chromategraphy and other separation and purification methods,30 compounds were separated and identified from the plant,including 1phenylpropanoids and 1 carbohydrates and 28 steroids according to their physical and chemical properties and HR-ESI-MS,¹H-NMR,¹³C-NMR,2D-NMR data analysis,they were identified as 3ξ-（1ξ-hydroxyethyl）-7-hydroxy-1-isobenzofuranone（1）,α-methyl aclepobioside（2）,Δ⁵-3β,8β,14β-trihydroxy-11α,12β-O-ditigloyl-pregnane（3）,Δ⁵-3β,8β,14β-trihydroxy-11α,12β-O-dibenzoyl-pregnane（4）,obcordata D（5）,obcordata I（6）,obcordata A（7）,obcordata J（8）,obcordata C（9）,obcordata K（10）,obcordata L（11）,obcordata G（12）,obcordata B（13）,choleata-5,20,24-trien-3β-ol（14）,3-O-[6-deoxy-3-O-methyl-β-allopyanosyl（1→4）-β-oleandropyranosyl]-5,6-dihydroge n-11α,12β-di-O-tigloyl-17β-m-arsdenin（15）,aspidatasin A（16）,aspidatasin B（17）,aspidataside A（18）,aspidataside B（19）,aspidataside F（20）,aspidataside C（21）,aspidataside D（22）,aspidataside E（23）,aspidataside G（24）,aspidataside H（25）,aspidataside I（26）,aspidataside J（27）,aspidataside K（28）,aspidataside L（29）,aspidataside M（30）.Compounds 8,10,11,16-30 are new compounds and compounds1-4,8,10,11,14-30 were isolated from the A.obcordata Hemsl.for the first time.The antitumor activities of pregnane glycosides isolated from A.obcordata Hemsl.against HL-60（human acute leukemia cells）,SW480（human colon cancer cells）and Huh-7（human liver cancer cells）were determined by MTT method.The results showed that compounds 18,19 and 21 had inhibitory activity on HL-60 cells,the IC₅₀ values are 13.2μg/ml,8.0μg/ml and 10.5μg/ml,respectively.Compound 18had different inhibitory activity on SW480 cells,the IC₅₀ values are 22.0μg/ml.Compounds 11 and 20 also had certain activity on Huh-7 cells.the IC₅₀ values are18.3μg/ml and 18.5μg/ml,respectively.Through the study on the chemical constituents and in vitro antitumor activity of Aspidopterys obcordata Hemsl,it is found that Aspidopterys obcordata Hemsl mainly contains pregnane glycosides,which have significant antitumor activity and may become potential antitumor drugs.
