The aging of the population has become one of the common problems concerned by the whole society.The elderly population is increasing rapidly year by year,followed by the obvious increase of senile diseases,which brings great pressure and challenges to the country,society,economy,culture and medical treatment.These senile diseases are caused by many factors,and the aging of the body is a common factor.Aging is an inevitable physiological process of the organism,but it is possible to delay the process of aging.Nematodes are selected as the main model organisms in the field of aging because they have the following advantages:(1)simple structure,short life cycle and easy cultivation;(2)transparent and easy observation of the body,easy statistical analysis of the number of offspring;(3)and human The genetic similarity reaches 60%-80%;(4)the experiment operation is simple.In this study,the model organisms Caenorhabditis elegans and P.dwarf elegans were used to study and analyze the alcohol extracts of Panax ginseng based on their anti-aging methods.This method mainly studies the effects of alcohol extracts of Panax spp.on physiological indicators such as the lifespan of nematodes and the experiment of nematode movement behavior.First,use the nematode life test to select the optimal concentration of the alcohol extract of Panax spp.(this experiment uses ginseng,American ginseng,and Panax notoginseng)to delay the life of the nematode,and then study the effect of the alcohol extract of the concentration of Panax spp.on other molecular indicators of the nematode.Ginseng,American ginseng,and Panax notoginseng may delay the lifespan of nematodes.The main research contents are as follows:1.The best experimental concentration of ethanol extract of Panax spp.was screened by the longevity test of the male worm of the dwarf P.The experimental results showed that the optimal concentration of ethanol extract of Panax spp.to delay the life of nematodes was 0.3 mg/m L for red ginseng,0.6 mg/m L for American ginseng,and 1.6mg/m L for Panax notoginseng.2.The life test results of ethanol extract of Panax ginseng against Caenorhabditis elegans showed that the average life span of the red ginseng(0.3MH)group was extended by 25.33%;the average life span of the 1.6mg/m L panax notoginseng(1.6MSC)group was extended by 17.33%;0.6 The average life span of the mg/m L American ginseng(0.6MXC)group was extended by 25.33%(P<0.05).In terms of extending the lifespan of nematodes,red ginseng is the best,followed by American ginseng,both of which are better than panax notoginseng.3.The movement behavior of the nematode gradually weakens with the gradual aging of the nematode.The results of this experiment show that the alcoholic extract of Panax ginseng can swing the head and the body of the nematodes from the 4th day of life.The alcoholic extract of Panax nematodes is higher than that of the blank group,and there are significant differences.On the 8th day,the 1.6M panax notoginseng group was basically paralyzed,while the nematodes in the 0.3M red ginseng group and the 0.6M American ginseng group were still active,and the 0.6M American ginseng group was better than the 0.3M red ginseng group.4.Under the condition of 40℃ heat stress,the resistance of the nematodes in the ethanol extract group of Panax ginseng was significantly improved.The average life span of the 0.3MH group,1.6MSC group,and 0.6MXC group was prolonged by 14.81%(P<0.01),25.93%(P<0.01),and 22.22%(P<0.01),respectively.Panax notoginseng prolongs the longest lifespan of nematodes,but comprehensive evaluation shows that American ginseng has the best heat-resistance ability,followed by red ginseng,which is better than Panax notoginseng.5.The oxidative stress test of the ethanol extract of Panax ginseng on nematodes shows that the ethanol extract of Panax ginseng has a significant protective effect on the damage of the two oxides(hydrogen peroxide and juglone)of the nematode,effectively prolonging the life of the nematode,and resisting the two The damage trend of these oxides is consistent.The 1.6M panax notoginseng group has the strongest protective effect on oxide damage,and the 0.6M American ginseng group and the 0.3M red ginseng group are almost the same.6.Comparing the molecular index determination of ethanol extract of Panax spp.on Caenorhabditis elegans and P.dwarf nematodes,experiments show that the molecular index trends of the two model nematodes are the same.Ethanol extract of Panax ginseng can significantly reduce the content of ROS in nematodes.Comprehensive evaluation shows that the 0.6MXC group is the best,followed by the 0.3MH group,which are better than the 1.6MSC group.The ethanol extract of Panax ginseng can effectively reduce the accumulation of lipofuscin in the nematodes,the nematodes in the 0.6MXC group are the most obvious,the 0.3MH group is the second,and the 1.6SC is the weakest.The ethanol extract of Panax ginseng significantly improves the activities of SOD and CAT enzymes in nematodes.Comprehensive evaluation,the alcohol extract of Panax ginseng has the best SOD activity in nematodes: the 0.6MXC group is the best,followed by the 0.3MH group,and both are better than the 1.6MSC group.The CAT activity of ethanol extract of Panax ginseng on nematodes in vivo: 0.3MH group and 0.6MXC group are almost the same,and both are better than 1.6MSC group.The alcohol extract of ginseng compared the molecular indicators of Caenorhabditis elegans and P.dwarf.Although there are differences,the overall trend of changes is the same.Therefore,P.dwarf can be used as a model organism for anti-aging drugs. |
