| Objective: Schisandra chinensis is the fruit of Schisandra chinensis,an evergreen shrubbery herb.It has the pharmacological effects of regulating the nervous system and improving the internal blood circulation of the human body.It is a kind of nourishing Chinese medicine that has been widely studied and applied in modern medicine in China.Acorus tatarinowii Schott belongs to Acorus tatarinowii Schott,which is the dried rhizome of Acorus tatarinowii Schott.Acorus tatarinowii Schott has pharmacological effects on improving human blood circulation and respiratory system,and protecting neurological function.It is used to effectively treat acute epilepsy,tinnitus,stroke and other diseases.Based on the treatment principle of traditional Chinese medicine ’ kidney deficiency and phlegm stasis ’ and the compatibility theory of traditional Chinese medicine,the pharmacodynamic material basis and mechanism of astringent Chinese medicine Schisandra chinensis and resuscitation Chinese medicine Acorus tatarinowii Schott in the treatment of Alzheimer ’ s disease were preliminarily revealed.In this study,we first used UPLC-Q-TOF/MS to determine the effective components of Sc-At ethanol extract.Secondly,according to network pharmacology,we found that the role of Sc-At in regulating AD was mainly related to 14 active compounds.These active compounds mainly act on 58 targets.Through these targets,sphingosine metabolism,arachidonic acid metabolism and cholinergic synapse metabolism are the main theoretical pathways of the drug.Finally,sphingosine,arachidonic acid and other lipid and amino acid metabolites were identified in the plasma non-targeted metabolomics of AD model rats,which verified the theoretical pathway of Sc-At and provided theoretical support for the treatment of AD with complex components of traditional Chinese medicine.Methods: In this study,we first searched the relevant network pharmacology literature and database to find its theoretical targets,and then analyzed the chemical constituents of the ethanol extract of schisandrinus acorus tatarinowii by UPLC-Q-TOF/MS.The prepared ethanol extract was intragastrically administered to rats with Alzheimer ’ s disease.After 10 weeks of intragastric administration,the contents of related components in the blood of rats were determined to study the therapeutic mechanism of schisandrinus acorus tatarinowii ethanol extract on Alzheimer ’ s disease rats.Results: 1.The components and targets of schisandra-corus calamus were determined :according to the principle of drug-like(OB ≥ 30 %,DL ≥ 0.18),14 effective components of schisandra-corus calamus were screened,68 targets for theoretical treatment of AD were enriched to 85 KEGG signaling pathways.2.Preliminary identification of schisandra-corus calamus compounds,preliminary characterization of schisandra 17 lignans,12 kinds of volatile oil,10 kinds of fatty acids,4 alkaloids,19 kinds of miscellaneous compounds,Acorus tatarionwii Schott 9 terpenes,6 kinds of flavonoids,4 kinds of phenylpropanoids,2 kinds of lignin,6 kinds of fatty acids,21 kinds of miscellaneous compounds.3.According to the relevant provisions of Chinese Pharmacopoeia given schisandra-corus calamus alcohol extract of Alzheimer ’s disease model rats can significantly shorten the time to find the target quadrant,increase the number of crossing the target quadrant,reduce the content of sphingosine phosphate,phenylalanine and other sphingolipids in rat plasma,increase the content of arachidonic acid,vitamins,and play a positive role in the treatment of Alzheimer ’s by affecting lipid and amino acid metabolism.Conciusion: In this study,the UPLC-Q-TOF method was established for the determination of alcohol extracts from Schisandra chinensis and Acorus tatarionwii Schott,and the experimental results showed that the prepared extracts had therapeutic effects on Alzheimer ’s disease,which provided a reference for the treatment of Alzheimer ’s disease with Schisandra chinensis and Acorus calamus. |
