Objective:At present, the results of many studies on ABCB1 gene polymorphism and the efficacy of antipsychotic drugs are not the same.Through meta-analysis,the data of multiple studies are combined to improve the statistical power of the original results, which can solve the inconsistency of the results and draw Reasonable conclusion.Method:We conducted a comprehensive literature search of the Pubmed、Embase、CENTRAL、CINAHL、Web of Science、Cochrane Library、CNKI、WANFANG、VIP、CBM to identify relevant case-control studies and cohort studies published before January 15,2021.The quality of the included studies was evaluated and data were extracted, and the data were analyzed using Stata statistical software.Two-class data were analyzed by the M-H method, and OR and its 95% CI were used as outcome indicators. The extracted Mean±SD was analyzed using the inverse variance method, and SMD and its 95% CI were used as the outcome indicators. For the above data, adjust the effect model according to the heterogeneity.Analyze the correlation between the SNP gene polymorphisms of ABCB1 and the efficacy of atypical antipsychotics.Results:(1)There were 7 articles included in this study, including 1052 patients.A total of 7 ABCB1 polymorphic genes were studied.Among them,rs1045642,rs2032582,and rs1128503 appeared in two or more studies,so they were included in the Meta analysis;while rs4444903,rs13233308,rs2235048,and rs12535512 only appeared in one study, whichcan not be included.(2)rs1128503(C1236T)wt: CC carrier: CT/TT.The results showed that: SNP rs1128503 genotype had no significant effect on the treatment effect of atypical antipsychotics(OR=1.41,95%CI[0.88,2.26]).(3)rs1045642(C3435T):wt: CC;carrier :CT/TT。The results show that: SNP rs1045642 genotype has no significant effect on the treatment effect of atypical antipsychotics(SMD=0.04,95%CI [-0.28,0.35],P=0.03).Two studies in this SNP use the number of response people/total number of people as the result, the conclusion of meta-analysis has not changed(OR=2.41,95%CI[1.30,4.47]).(4)rs2032582(G2677T/A):wt: GG;carrier:GT/TT/GA/AA,The results showed that: SNP rs2032582 genotype had no significant effect on the treatment of atypical antipsychotics(SMD=0.02,95%CI[-0.18,0.22])Conclusion:There is no significant correlation between ABCB1 gene polymorphism and theclinical efficacyof atypical antipsychotics.
