Objective: To reveal the expression profile of circ RNAs in the peripheral venous blood of children with biliary atresia,and to explore the role of differentially expressed circ RNAs in the occurrence and development of biliary atresia,so as to provide a new direction for the study of biliary atresia.Methods:This study included 3 cases of biliary atresia(BA)and 3 cases of Cholangiectasia(CC),and high-throughput RNA sequencing technology was used to detect and compare the circ RNA expression profiles in whole blood samples of peripheral venous blood of 6 cases.By analyzing the sequencing results,q PCR was used to expand the sample size of some circ RNAs with significant differences in expression.Results:According to high-throughput sequencing of the whole transcriptome,a total of55,516 circ RNAs in the biliary atresia group were differentially expressed from those in the cholangiectasis group.To log2 fold change | > 1.0 and adj.P < 0.05 screening,there are 291 circular RNA expressed in the differences between the two groups have statistical difference.Compared with the control group,there were 169 circ RNAs with increased expression and 122 circ RNAs with decreased expression in the BA group.GO analysis suggested that the differentially expressed circ RNAs may be involved in the negative regulation of apoptosis in mature B cells and the positive regulation of cytokine-mediated signaling pathways.KEGG analysis suggested that differentially expressed circ RNAs play roles in lysosome,amoebiasis,ECM-receptor interaction and malaria signaling pathways.The five circ RNAs with significantly high expression were verified by expanding the sample size,and it was found that hsa_circ_0081144,hsa_circ_0044556and hsa_circ_0058738 were significantly higher in the BA group than in the CC group,and the difference between the two groups was statistically significant(P<0.0001).Conclusion:Hsa_circ_0081144,hsa_circ_0044556 and hsa_circ_0058738 are up-regulated in BA peripheral blood,and these three circ RNAs may play a role in the occurrence and development of biliary atresia.
