| Objective: Micro RNA plays an important biological role in all aspects of tumorigenesis and development.Previous research showed that the level of miR-21 was significantly increased in many cancers,including Medulloblastoma.However,the specific mechanism of miR-21 in medulloblastoma has not been reported.Therefore,this study aims to explore the role of miR-21 on medulloblastoma DAOY cells and its effect on related signal pathways.Method: MiR-21 inhibitor,miR-21 inhibitor NC and miR-21 mimic were transfected into DAOY cells by liposome TM2000.After 48 hours,the expression of miR-21 was evaluated by q RT-PCR method;the proliferation of cells in different transfection groups was verified and detected by CCK8 method and plate cell clone formation assay;the necrosis and apoptosis of cells in different transfection groups were detected by Annexin V-FITC assay;the invasion and migration ability of cells in different transfection groups were verified and detected by Transwell test and cell scratch test.At the same time,Western blot was used to detect the activation of PTEN/PI3K/AKT in different transfection groups,and the effect of miR-21 on PI3K/AKT signal pathway was observed.Results: Compared with inhibitor NC group,the proliferation,cell migration and invasiveness of DAOY cells transfected with miR-21 mimic were increased,while apoptosis was significantly decreased.After miR-21 inhibitor transfection,the proliferation,migration and invasiveness of DAOY cells were significantly decreased,while apoptosis was significantly increased.In DAOY cells transfected with miR-21 mimic,the expression of PI3 K and AKT increased significantly,while the protein levels of PTEN,Caspase-3 and Caspase-9 decreased significantly.At the same time,in DAOY cells transfected with miR-21 inhibitor,the expression of PI3 K and AKT decreased,while the protein levels of PTEN,Caspase-3 and Caspase-9 increased significantly.Conclusion:1.MiR-21 is highly expressed in medulloblastoma,and can promote the proliferation,infiltration,migration and invasion of medulloblastoma cells,and reduce the apoptosis of medulloblastoma cells.2.MiR-21 in medulloblastoma promotes the proliferation,infiltration,migration and invasion of medulloblastoma by regulating the expression of PI3K/AKT signal pathway. |
