Objective: A total of 1.6 million people are diagnosed with oral squamous cell carcinoma every year.Due to its characteristics of easy lymph node metastasis and poor prognosis,the 5-year overall survival rate is only about 63%.High-throughput sequencing showed that circ RNAs were abundant in oral squamous cell carcinoma cells,and most of them were formed by cyclization of exons encoding proteins.In this study,the expression level of hsa_circ_0009682 in oral squamous cell carcinoma was verified,the clinicopathological data of hsa_circ_0009682 was collected,and the function enrichment analysis of hsa_circ_0009682 was conducted through big data to explore the clinical significance of hsa_circ_0009682 in tumor diagnosis,and to provide a new potential marker for tumor diagnosis and treatment.Methods: According to the previous analysis of circ RNAs high-throughput sequencing results and the literature that we have consulted,we selected a significantly differential expressed circ RNA which has not been studied in oral squamous cell carcinoma as a target circ RNA.The relative expression level of hsa_circ_0009682 was detected in 60 pairs of OSCC tissue samples and corresponding adjacent normal tissues(AHTs),cell lines(CAL27,SCC9 and its corresponding human normal oral keratinocytes(HOK))by q RT-PCR.Additionally,we analyzed the relationship between the expression level and clinicopathological characteristics.Through DO,GO,KEGG and Reactome functional enrichment analysis and bioinformatics analysis were conducted to further understand the potential functions of hsa_circ_0009682 and the targeting relationship of hsa_circ_0009682-mi RNAs-mRNAs,Finally,the interaction analysis network was obtained.Diagnostic value of hsa_circ_0009682 in OSCC was assessed by the ROC curve.Results: The expression level of hsa_circ_0009682 in OSCC tissue samples and OSCC cell lines was discovered to be significantly down regulated compared with the AHTs and HOK.Moreover,hsa_circ_0009682 expression level was significantly related to TNM stage and tumor size.Hsa_circ_0009682-mi RNAs-mRNAs network analysis,GO,DO,KEGG and Reactome analyses all demonstrated that hsa_circ_0009682 related to cancer progression to a certain extent.The area under the ROC curve was 0.623 which has good diagnostic value.Conclusion: The expression of Hsa_circ_0009682 in oral squamous cell carcinoma tissues and cells was significantly lower than that of the control group,and the patients with low relative expression level of OSCC were more likely to have later stage of TMN and larger tumor volume,and there was a potential important biomedical mechanism in the tumor.Therefore,Hsa_circ_0009682 can be used as an unstudied potential marker of OSCC,which has a positive role in the early diagnosis rate of OSCC.
