Objective:Using network pharmacological research strategy,the mechanism of action of Shenfukang Ⅱ capsule in the treatment of renal interstitial fibrosis was investigated.Methods:The potential active ingredients in Shenfukang Ⅱ capsule were collected by the TCMIP and the TCMSP databases.Based on the chemical similarity principle,the target of potential active ingredients was predicted by using the TCMIP.Renal interstitial fibrosis related genes were retrieved from Disge Net,ETCM and Gene Cards databases and mapped to component targets,the mapping results were imported into David database for biological function analysis.Cytoscape3.7.0 software was used to construct and analyze the network of "Herb-components-targets" and "Herb-components-targets-pathways" of Shenfukang Ⅱ capsule.The common targets were imported into the STRING database for PPI analysis,the result was imported into Cytoscapersoftware for visualization and analysis.Finally,the mechanism of Shenfukang Ⅱ capsule in the treatment of RIF was analyzed by combining the results of "H-C-T","F-H-C-T-P" network analysis and PPI interaction results.Results:Shenfukang Ⅱ capsule contains a total of 198 potential active ingredients,1011Ingredients-targets were obtained by reverse prediction,and 226 potential anti-RIF targets of Shenfukang Ⅱ capsule were obtained by mapping with 2117 RIF-related genes.A variety of KEGG pathways were obtained by enrichment of these targets.In terms of GO analysis,BP,CC and MF all involve multiple items."H-C-T" and "F-H-C-T-P" networks were analyzed by cytoscape.then 37 key components and 36 core targets were obtained.Finally,the network analysis of "F-H-C-T-P" was integrated to find out that quercetin,kaempferol,palmitic acid,ursolic acid,oleanolic acid,adenosine and other components might be the main anti-RIF active components.Combined with the results of PPI analysis,TNF,NFKB1,MAPK3,Casp3,AR and NR3C1 may be the main anti-RIF targets of Shenfukang II.Conclusion:The role of Shenfukang Ⅱ capsule in the treatment of RIF lies in the characteristics of multi-component,multi-target and multi-channel.Specifically,the pharmacodynamic components in Shenfukang Ⅱ may regulate PI3K-Akt signaling pathway,HIF-1 signaling pathway and other pathways by intervening TNF,NFKB1,MAPK3,CASP3,AR,NR3C1 and other targets.Intervention of inflammatory response,oxidative stress,regulation of blood pressure,blood glucose and blood lipid or other processes to play a role in the treatment of renal interstitial fibrosis.
