| Objective To detect disparate number of m RNA and the Long non-coding RNA(LncRNA)in intervertebral discs of patients with spinal tuberculosis(STB)and spinal degeneration by microarray,and to screen LncRNA with potential as a diagnostic marker.Methods From 9.2018 to 12.2020,the intervertebral disc came from 28 patients with STB and 28 patients with spinal degeneration who got treatments in General Hospital of Ningxia Medical University.Extraction of total RNA of intervertebral disc and then check quality of them.Three samples of qualified RNA quality were selected and detected by Arraystar human LncRNAV5.0 chip.The m RNA and LncRNA with Fold Change≥ 2 and P < 0.05 were screened out.Among them,m RNA adjacent to LncRNA were regarded as cis-target genes,the proteins that LncRNA may bind were regarded as trans-target proteins,and the gene function annotation analysis was carried out respectively to explore the possible function of m RNA and LncRNA in the pathogenesis of STB.The LncRNA whose cis-target genes may lead to STB were selected.We did RT-PCR to detect the number of the lncRNA mentioned above in the other 25 samples,and further screening the markers that may be related to the diagnosis of STB.Results Based on the chip data,a total of 444 differentially expressed LncRNA(up-regulated 398,down-regulated 46)and 186 m RNA(up-regulated 107,down-regulated 79)were screened.The gene function annotation analysis showed that cis-genes can regulating biological processes.Just like gene expression,leukocyte adhesion and migration,immune response,endocytosis and redox reaction,as well as in the transmission of NF-κ B,endocytosis and metabolic pathway.The trans-proteins can also regulating gene expression;The m RNA can affect biological processes too.Just like gene expression,apoptosis,bacterial defense,inflammation,immune,and the transmission of signaling pathways such as endocytosis and metabolic pathway.RT-PCR showed that LINC01128-201 and DNALI1-205 were differentially expressed in patients with STB,which was same as the microarray data.Conclusions 1.There are disparate number of LncRNA and m RNA in the intervertebral disc between STB patients and degenerative patients.2.The above-mentioned LncRNA may regulate the amount of cis-target gene by binding with trans-target protein,and then regulate the function of endocytosis,apoptosis,inflammation and immune response through TGF-β,NF-κB signal pathway,regulate the immune defense ability of host to MTB and participate in the pathogenesis of STB.3.Linc01128-201 and DNALI1-205 are significantly low expressed in the intervertebral discs of patients with STB.Their cis-target genes ISG15,HES4 and SNIP1 may lead to the onset of STB and have the potential to become diagnostic markers. |
