Objective:Polyether ether ketone(Peek)material,which has stable chemical structure,elastic modulus similar to bone,superior mechanical strength but no biological toxicity,is currently one of the commonly used materials for orthopedic implants.However,the bio-inertness is poor in integrating with the surrounding bone tissue,making its clinical application limited.The strontium Eucommia ulmoides polysaccharides(Eup-Sr)is a new type of biologically active complex,which has been proved to possess immune-modulating ability.Therefore,in order to improve the biocompatibility and biological function of pure Peek,it is modified by surface treatment to attain Dppeek,on the surface of which Eup-Sr is introduced to produce Eup-Sr/peek.In this study the biocompatibility and biological function of Eup-Sr/peek is analyzed to provide theoretical basics for future study and clinical application of modified Peek materials as a new ideal orthopedic implant.Methods: The experiment is divided into three stages: material preparation,characterization and in-vitro cell experiment.Firstly,the Eup-Sr and the functional surface of Peek with polydopamine(Dppeek)are prepared.Then the Eup-Sr solutions are prepared with concentrations of 10μg/ml,50μg/ml,100μg/ml,and 500μg/ml respectively,and used to modify the functional surface of Dppeek in order to get the composite material Eup-Sr/peek.Secondly,the surface morphology,surface elemental composition after modification,and surface contact angle of the Peek,Dppeek and Eup-Sr/peek are detected with scanning electron microscope and X-ray photoelectron spectroscopy(XPS).Thirdly,Eup-Sr/peek were co-cultured with MC3T3-E1 cells and their proliferation ability was detected by MTT method.Osteogenic and inflammatory factors were detected with q RT-PCR and the alkaline phosphatase activity was detected by alkaline phosphatase staining.Results:The experiment successfully integrated the prepared Eup-Sr on the surface of Dppeek that was treated with concentrated sulfuric acid sulfonation,water bath heating,plasma treatment and polydopamine surface coating to obtain Eup-Sr/peek composite material.Compared with the Peek,the surface of the Eup-Sr/peek and Dppeek showed a three-dimensional nano network structure and increased surface roughness under the scanning electron microscope.The XPS showed that the Sr3p3 and Sr3d5 characteristic peaks were detected on the surface of the Eup-Sr/peek composite material,indicating that the chemical composition had changed significantly and new chemical element had been introduced on the surface of Dppeek.The surface contact angle detection showed that the hydrophilicity of Eup-Sr/peek greatly improved compared to that of the Peek surface(p<0.001).In the in-vitro cell experiment,the cell viability detection with MTT on the second day showed that the Eup-Sr/peek composite material exhibited superior performance in promoting the proliferation of MC3T3-E1 cells(p<0.001).And the result on the fourth day showed that expect the Eup-Sr/peek composite material in the 500μg/ml concentration group,orther groups showed the superior performance in promoting the proliferation of MC3T3-E1 cells(p < 0.001).However,the 500μg/ml concentration of the Eup-Sr/peek composite material was cytotoxic(p<0.001).The q RT-PCR result showed that the expression of ALP,Col1α1,RUNX2 were significantly increased(p<0.01)and CR expression was significantly decreased(p<0.01)under the effect of the Eup-Sr/peek composite material.At the same time,the expression of inflammatory genes including interleukin-1β and interleukin-18 was significantly decreased(p<0.01).The alkaline phosphatase staining indicated that the Eup-Sr/peek Group had higher alkaline phosphatase activity than the Peek and the Dppeek Group.Conclusions: In this study,the Eup-Sr/peek new material was successfully prepared from the preparation and integration of Eup-Sr and Dppeek.With Sr successfully introduced into Eup-Sr/peek,it was found that Eup-Sr/peek has a three-dimensional nano network structure and high hydrophilicity.At the aspect of function,Eup-Sr/peek has good biocompatibility,pro-osteogenic ability and anti-inflammatory effect.This study provides experimental basic support for the clinical application of modified Peek materials as a new ideal orthopedic implant.
