| Objective : Lung cancer is the highest incidence of cancer in the world after female breast cancer,and also the highest mortality of cancer.LUAD is the main pathological type of lung cancer.most of the patients are already in the middle and advanced stage when diagnosed,which has caused a great burden on families and society.SFXN1 plays the role of mitochondrial serine transporter in carbon unit metabolism,is closely related to the energy metabolism process of tumor.At present,the expression and the role of SFNX1 in LUAD are still unclear.Therefore,this study aims to explore the clinical expression and preliminary biological function of SFXN1 in LUAD,to further explore the molecular mechanism of LUAD pathogenesis,so as to provide experimental evidence for clinical diagnosis,prognosis assessment and molecular targeted therapy of LUAD.Methods: First,the expression data of SFXN1 in lung adenocarcinoma were searched and downloaded by The Cancer Genome Atlas(TCGA)public dataset.The expression difference of SFXN1 in LUAD and corresponding paracancerous normal tissues was compared and analyzed,and the effect of SFXN1 expression level on disease-free survival of LUAD patients was further analyzed using a variety of clinicopathological parameters.Then,at the clinical level,70 cases of LUAD patients were collected for surgical resection of cancer tissues and corresponding paracancer tissues.q RT-PCR and WB was used to detect the m RNA and the protein expression level of SFXN1 in the cancer and paracancer tissues.Statistical methods were used to calculate the expression level of SFXN1 in cancer tissues and corresponding paracancer tissues,then analyze the relationship between SFXN1 and the tumor’s degree of differentiation,lymph metastasis,clinical stage and other clinicopathological characteristics.Lentiviral overexpression vectors were constructed to overexpress SFXN1 in LUAD cell lines A549 and H1299,and the effects of SFXN1 on the biological functions of LUAD were investigated by cell proliferation,migration,invasion and apoptosis experiments.Results:1.By analyzing the results of the TCGA database,it was found that the expression of SFXN1 in LUAD was significantly increased compared with the adjacent normal tissues(P<0.05),and high SFXN1 expression level,Stage is an independent risk factor for poor prognosis in LUAD patients.2.The results of the q RT-PCR and WB showed that SFXN1 expression in LUAD tissues was significantly higher than the adjacent normal tissues(P<0.05),and it was related to TNM(P<0.05)stage and the N stage,and had no significant correlation with gender,age,tumor size,smoking or not(P>0.05).3.The up-regulation of SFXN1 expression can significantly enhance the proliferation,migration and invasion ability of LUAD cell lines A549 and H1299,and inhibit the apoptosis of A549 and H1299.Conclusion:1.The expression of SFXN1 was significantly increased in LUAD tissues,and was closely related to TNM stage and N stage.2.In vitro cell experiments showed that SFXN1 could enhance the proliferation,migration and invasion of LUAD cells and inhibit their apoptosis.These results suggest that SFXN1 may play a role in the occurrence and development of LUAD. |
