Objective: Thyroid carcinoma is a common malignant tumor in clinical practice,which affects the life and health of patients.It is of great significance to study the molecular level characteristics of thyroid carcinoma,study its pathogenesis and find an effective treatment.Therefore,in this study,we detected the content of sodium iodide symporter(NIS)protein in the cancer tissues of thyroid cancer and adjacent thyroid tissues to clarify the correlation between the expression of NIS protein and thyroid cancer and provide a theoretical basis and guidance for the diagnosis,treatment,and prognosis of thyroid cancer in clinical practice.Methods: A total of 50 patients(27 males and 23 females;aged 9-80 years,65 years)who met the diagnostic criteria of thyroid cancer and had complete clinicopathologic data were selected from our hospital from October 2019 to August 2020.Part of the resected thyroid cancer tissue was taken during surgery,and then the thyroid tissue of more than appropriate cases was taken as a control.The expression differences of NIS protein in the two tissues were detected by immunohistochemistry and Western blot,and then the relative expression contents of NIS mRNA in the two tissues were detected by real-time PCR.Finally,the expression differences of NIS protein and mRNA in thyroid cancer tissues and tissues were analyzed to assess the correlation between NIS protein and thyroid cancer.Results: The immunohistochemical staining products of NIS in thyroid carcinoma were mainly localized in the cytoplasm,and showed diffuse and tan dark staining;the positive products of NIS in thyroid adjacent tissues were localized in the cell membrane and cytoplasm,showing yellowish and scattered staining.1.Immunohistochemical results showed that: among 50 patients with thyroid cancer,no negative expression of NIS was observed in the tissues adjacent to thyroid cancer and thyroid cancer;49 patients(98%)had weak positive expression of NIS protein in the tissues adjacent to thyroid cancer and 9 patients(18%)had weak positive expression of NIS protein in thyroid cancer,and the weak positive expression rate of NIS in thyroid cancer was significantly lower than that in the tissues adjacent to thyroid cancer,and the difference had statistical significance(χ2 = 65.68,P < 0.05);1 patient(2%)had positive expression of NIS protein in the tissues adjacent to thyroid cancer and 14 patients(28%)had positive expression of NIS protein in the tissues adjacent to thyroid cancer,and the positive expression rate of NIS in thyroid cancer was significantly higher than that in the tissues adjacent to thyroid cancer,and the difference had statistical significance(χ2 = 13.26,P < 0.05);0 patient(0%)had strong positive expression of NIS protein in the tissues adjacent to thyroid cancer,and 27 patients(54%)had strong positive expression of NIS protein in the tissues adjacent to thyroid cancer.2,Western Bolt test results showed that thyroid cancer tissues(1.71 ±0.48 mg/m L)were higher than thyroid cancer adjacent tissues(0.48 ± 0.19 mg/m L),and the difference was statistically significant(P < 0.05).3,the results of real-time PCR detection of NIS mRNA showed that thyroid cancer tissues(0.87 ± 0.09)were higher than thyroid cancer adjacent tissues(0.27 ± 0.12),and the difference was statistically significant(P < 0.05).Conclusion: 1.The weak positive rate of NIS protein expression in thyroid carcinoma is significantly lower than that in adjacent tissues of thyroid carcinoma.The positive rate and strong positive rate of NIS protein expression in thyroid carcinoma are significantly higher than those in adjacent tissues of thyroid carcinoma.2.the relative expression of NIS protein and mRNA was higher in thyroid cancer tissues than in adjacent thyroid cancer tissues.3.there was a correlation between the expression of NIS protein and thyroid cancer. |
