Objective: The purpose of this study is to explore gene methylation of aggressive behavior in pediatric bipolar disorder with depressive episode from epigenetics.Methods:1.First we collect 10 pediatric bipolar disorder patients with depressive episode,5 cases from aggressive behavior group,5 cases from non-aggressive behavior group in clinical psychology department of the people’s hospital of Xinjiang Uygur Autonomous Region.5 milliliters of venous blood were extracted on an empty stomach and DNA was extracted.The Illumina 850 K methylation chip and Genome Studio software were used to screen the different methylation genes which related to the aggressive behavior in pediatric bipolar disorder with depressive episode.We used the online Database David(The Database for Annotation,Visualization and Integrated Discovery)for Gene Ontology analysis and Pathway analysis to understand the functions and participating pathways of differential methylated genes between the two groups,and DPYSL2 and ABI3 BP genes were identified as candidate genes for pyrosequencing.2.We collect 45 pediatric bipolar disorder patients with depressive episode,16 cases from aggressive behavior group,29 cases from non-aggressive behavior group in clinical psychology department of the people’s hospital of Xinjiang Uygur Autonomous Region.5 milliliters of venous blood were extracted on an empty stomach and DNA was extracted.Pyrosequencing was used to verify DPYSL2 and ABI3BP’s methylation degree between the two groups.IBM SPSS Statistics 25 software was used for statistical analysis of the experimental data.Results:1.The gender,age,onset age,course of disease,family history,and HAMD scores of the two groups did not differ(P=0.988,0.100,0.906,0.428,1.000,0.087).2.There were 172 hypermethylation loci,148 hypomethylation loci,89 hypermethylation genes such as DPYSL2,and 78 hypomethylation genes such as ABI3 BP,which were different between the two groups of samples.The peak value of methylation β in aggressive behavior group was about 0.625,which were about0.40 and 0.85,respectively in non-aggressive behavior group.3.Pathway analysis showed that ST3GAL2,GALNT18,GALNT9 methylation genes were involved in mucin-type O-glycan biosynthesis Pathway.4.The methylation rates of DPYSL2 gene in the two groups were 27.438±17.874% and 27.345±18.832%,respectively,with no statistical significance(P=0.113).The methylation rates of ABI3 BP gene in the two groups were 84.188±25.761% and 78.586±23.567%,respectively.The difference was not statistically significant,too(P=0.987).Conclusion:1.There were DPYSL2 methylation gene and ABI3 BP methylation gene between the two groups;2.There was no significant correlation between aggressive behavior in pediatric bipolar disorder with depressive episode and DPYSL2 and ABI3 BP genes.
