Objective:To investigate the diagnostic value of serum miR-129 and miR-205 in Chronic kidney disease(CKD)patients with vascular calcification(VC).Methods:Fifty-four patients with CKD admitted to our hospital from June 2020 to January 2021 were selected,and the patients were divided into non-calcification group(CACS=0)and calcification group(CACS>0)according to the results of Coronary Artery Calcification Score(CACS),of which 21 cases were in the non-calcification group and 33 cases in the calcification group.Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR)was used to detect the relative expressions of miR-129,miR-205,Runx2 in serum.Two independent samples t-test or non-parametric test were used to compare the differences in clinical biochemical indexes,miR-129,miR-205 and Runx2 expression in serum between the two groups.Spearman test was used to analyze the correlation between miR-129,miR-205and Runx2 in serum and clinical biochemical indexes and vascular calcification.Receiver operating curve(ROC)was used to analyze the diagnostic value of serum miR-129,miR-205 and Runx2 alone and in combination in the diagnosis of vascular calcification in patients with chronic kidney disease.Results:1.Age,systolic blood pressure and serum phosphorus levels in calcification group were significantly higher than those in non-calcification group(P<0.05),and serum calcium and albumin levels were significantly lower than those in non-calcification group,with statistical significance(P<0.05).2.The expression levels of miR-129 and miR-205 in the calcification group were lower than those in the non-calcification group,and the expression level of Runx2 was higher than that in the non-calcification group,with statistical significance(P<0.05).3.Spearman correlation analysis showed that the expressions of miR-129 and miR-205 in serum were positively correlated with serum calcium and albumin levels,and negatively correlated with age and vascular calcification level(P<0.05).The expression of Runx2 in serum was positively correlated with vascular calcification level(P<0.05).4.ROC curve showed that the Area Under ROC Curve(AUC)of serum miR-129,miR-205 and Runx2 when vascular calcification was diagnosed alone was 0.763,0.831 and 0.666,respectively.The AUC of miR129+205=0.856,miR-129+Runx2=0.798,miR-205+Runx2=0.823 when combined detection,respectively.The AUC of the three combined detection was 0.851.Among them,the AUC corresponding to miR-129 and miR-205 combined detection of VC was the largest,which had better diagnostic value.Conclusions:1.The expression of miR-129 and miR-205 is low and Runx2 is high in serum of CKD patients with vascular calcification.2.Mi R-129,miR-205 and Runx2 are potential diagnostic markers for vascular calcification in patients with CKD.Mi R-205 has a better diagnostic efficacy when detecting vascular calcification with a single indicator,and miR-129 and miR-205 have the highest diagnostic efficacy when detecting vascular calcification with a combination of multiple indicators.
