ObjectiveThis study aimed to investigate the clinical significance and related pathway of ALDOA expression in BUC.MethodsThe expression information and clinicopathological data of ALDOA in bladder cancer and normal bladder tissues were obtained from TCGA database,and the relationship between ALDOA and clinical pathological features such as clinical stage,metastasis,histological grade and prognosis of BUC was analyzed.GSEA(Gene Set Enrichment Analysis)was used to predict the related pathways and mechanisms of ALDOA.45 patients with urothelial carcinoma of bladder were collected from April2019 to June 2020 in our hospital,and the expression of ALDOA was detected by immunohistochemical staining and Western blotting,including 45 normal tissues adjacent to the cancer as control.To compare the difference of ALDOA expression between urothelial carcinoma of bladder and normal tissue,and to further verify the correlation between ALDOA expression and clinicopathological parameters and its effect on prognosis.ResultsThe results of TCGA data analysis showed that the expression of ALDOA in bladder urothelial carcinoma was 16.07-16.23,with a median of 16.13,which was significantly higher than that in normal tissues(15.40-15.93,15.68),and the result was significant(P<0.014).In the analysis of the relationship between the expression level of ALDOA and clinicopathological features.There was a significant correlation between the expression of ALDOA and the histological grade of BUC(P= 0.023).Kaplan-Meier survival analysis showed that the high expression of ALDOA in patients with bladder urothelial carcinoma was significantly correlated with poor prognosis.In further clinical verification,the immunohistochemical results showed that the staining intensity of BUC was higher,which was lower in normal tissues,and the average optical density was 0.027±0.023 and 0.0029±0.00064 respectively.The difference was statistically significant.The average optical density in high grade bladder cancer was higher than that in low grade bladder cancer,and the difference was statistically significant too.In the further quantitative analysis of western blot experiment,the gray value of bladder urothelial cancer tissue is higher than that of normal tissue adjacent to cancer,which proves the high expression of ALDOA in tumor tissue again.Finally,GSEA analysis showed that there were glycolysis、oxidative phosphorylation、hypoxia pathway and Myc gene sets related to cell metabolism and proliferation in samples with high expression of ALDOA.ConclusionsThe expression of ALDOA is up-regulated in BUC,which is closely related to the histological grade of BUC.The high expression of ALDOA is a poor prognostic factor.ALDOA can be used as a biomarker for the diagnosis and prognosis of BUC,and may be a potential target of treatment.
