Chitosan(CTS)is the only basic polysaccharide in nature,which is a macromolecular polymer and has good solubility in an acidic solution.Chito-oligosaccharide(COS),a degradation product of CTS,is soluble.CTS and COS have good biological activity,such as antibacterial,antioxidant,anti-tumor,anti-inflammatory effects,and wide application in the field of pharmaceutical and health care products.However,the exact quantitative quantity of CTS and COS is still a problem.Therefore,it is especially important to establish CTS and COS accurate quantitative quantities.CTS and COS have the same molecular unit structure.The difference between of them lies in the difference of molecular chain length and molecular weight.According to the literatures,the detection methods of them were similar.However,due to the difference in molecular weight,the microscopic morphology,interface characteristics and interaction between the substances may be very different.There are certain differences in the spectral characteristics of the detection system,which leads to differences in the sensitivity and anti-interference ability of the detection methods.In this study,three different types of systems were investigated,and the RRS intensity was used as the main response signal.In the same detection system,the RRS was combined with other technique to characterize the microscopic characterization of the detection system,comprehensively.Further in-depth discussion of the quantitative analysis mechanism of CTS and COS also provides an important reference for the in-depth study of RRS quantitative methods.In the three detection systems,each experimental parameter was optimized and the effect of ionic strength,sensitivity,precision and recovery of each method on the quantitative analysis were investigated.At the same time,combined with sample pre-processing methods,the three methods were applied to the determination and analysis of actual health products on the market.The results shown that the established three types of detection methods were simple,sensitive,accurate and highly selective,and suitable for the accurate quantification of CTS and COS in health care products.The specific research contents and results of this article were as follows:1.Comparative study on quantitative analysis of chitosan and chitooligosaccharides by enhanced resonance Rayleigh scattering with allure red probeThis study used the protonation of amino groups in CTS and COS under weak acid conditions,combined with the anionic azo dye Allure Red(AR)through ion association,and further enhanced the sensitization by surfactants,establishing a new quantitative analysis of CTS and COS method.Combining transmission electron microscopy(TEM),RRS spectra and UVVis,the microscopic effects of the three method were analyzed,and the difference between the RRS method in detecting COS and CTS was further studied.It was found by comparison that,due to the difference in the molecular weight of COS and CTS,the main forces in the two systems were different.Within a certain range,the RRS intensity was linearly with the increase of COS or CTS concentration,which served as a quantitative basis.2.Comparative study on quantitative analysis of chitosan and chitooligosaccharides by Resonance Rayleigh scattering quenching with Eosin Y as probeIn this study,based on the quenching of CTS and COS on the RRS intensity of Eosin Y,a new method for quantitative analysis of CTS and COS was established.In this study,the RRS was combined with the ζ potential,UV-Vis and other microscopic characterizations to comprehensively analyze the interaction between the substances in the complex,and the quenching principle of the RRS intensity of Eosin Y by CTS and COS was analyzed.Through a series of experimental results and comparing the two systems of CTS and COS,it was found that the difference between the two detection systems was mainly reflected in the difference in sensitivity.Within a certain range of the two detection systems,the RRS intensity decreased linearly with the increase of COS or CTS concentration,which served as a quantitative basis.3.Comparative study on quantitative analysis of chitosan and chitooligosaccharides by direct blue 86 coordination compoundsThis study uses the empty orbital provided by the incompletely coordinated copper atom in direct blue 86 to coordinate with the nitrogen on the unprotonated amino group on CTS and COS to form a supramolecular complex with a dense structure and a large rigid plane.Established a new method for quantitative analysis of CTS and COS based on the highly sensitive RRS method of coordination interaction.Through TEM,ζ potential,RRS and UV-Vis,the interaction between the substances in the complex and the coordination mechanism of direct blue 86 with CTS and COS was analyzed.The difference between CTS and COS detection systems was also compared.The comparison found that the difference between the two detection systems is mainly reflected in the difference in force,antiinterference ability and sensitivity.Within a certain range of the two detection systems,the RRS intensity increased linearly with the increase of the concentration of CTS or COS,which served as a quantitative basis.
