| The thesis includes two sections:(40).Chemical constituents and anti-inflammatory activity of roots of Oxybaphus himalaicus;(40)(40).Structure elucidation of two new compounds of Securidaca inappendiculata.Ⅰ.Chemical constituents and anti-inflammatory activity of Oxybaphus himalaicusOxybaphus himalaicus Edgew.is an annual herbaceous plant which belongs to genus Oxybaphus(family Nyctaginaceae).It is mainly distributed in Shanxi,Gansu,Sichuan,Yunnan and Tibet.The roots of the plant are used as a traditional Tibetan medicine,which exhibits the medical effects of kidney warming,aphrodisiac,diuretic,urinary calculus removing and damp heat relieving,and are used to treat arthralgia,kidney weak cold,edema,hypogastralgia,bladder stones,virus infection and various venereal diseases.As a continuing investigation of chemical constituents and anti-inflammatory activities of the roots of O.himalaicus,fourteen compounds have been isolated from the n-butanol extract by various chromatographic methods including Sephadex LH-20gel chromatography,MPLC and preparative HPLC.According to the data of spectral analysis(MS,IR,UV and NMR),six rotenoids,borravinone A,B,D,F,K,and X;one isoflavanone:dihydroformononetin;one pterocarpin:medicarpin;and one carboxylic ester:dibutylphthalate were identified.The anti-inflammatory activities of isolated compounds were evaluated.Among them,borravinone A and D exhibited the most potent inhibitory activities(IC50 8.56 and 1.02μM,respectively)to NO release.The results showed that these compounds may have further research and development value and provided scientific basis for elucidating the traditional use of the medicine.Ⅱ.Structure elucidation of two compounds from roots of Securidaca inappendiculataS.inappendiculata Hassk is a climbing plant which belongs to genus Securidaca,family Polygalaceae and is mainly distributed in Yunnan,Guangdong,Hainan and Guangxi of China and tropical regions of southeast Asia.There are approximately 80species in this genus,which are mainly distributed in tropical America,with a few species in tropical Asia and Africa.The medical parts of the plants are the stems and roots,which are bitter in taste and mild cold in nature.The medical efficacy includes activating blood circulation,dispersing blood stasis,relieving swelling,clearing heat and diuresis,and are used for injuries caused by bruising,rheumatism,bone pain and lumbar muscle fatigue.It is recorded in the Chinese Pharmacopoeia of 2015 as being one of the main medicines in Yulin Brand Bone-setting Water,which is applied to treat fall injury,fracture,dislocation and elimination of sports fatigue.Fourteen compounds were reported by Wang from the roots of S.inappendiculata in our lab,including two new hemiterpene glycosides,1 benzyl benzoate and 1 lignan glycoside.Other 10 known compounds,including lignan glycosides and tannins.The screening results of cytotoxicity in vitro shows that all the new compounds showed cytotoxicity.As the continuing experiment of the same fraction,by using the combination of various isolation methods,such as MPLC,HPLC and identification methods,such as MS,1D,2D NMR,two new compounds(7R,8R,8′S)-glaberide I 4-O-β-D-glucopyranosyl-(1?→4″)-O-β-D-glucopyranoside(SEC 20)and 1-O-1,5-anhydro-glucitol-2-methene-butyrate-4-O-β-D-glucopyranoside(SEC 21)have isolated and identified. |
