Objective To study the effects of bufalin on cell proliferation,apoptosis and cell cycle of human hepatocellular carcinoma HepG2 cells in vitro.To study the radiosensitization of bufalin on human hepatocellular carcinoma HepG2 cells To explore the feasibility of the application of buflin and radiosensitization of hepatocellular carcinoma,and to provide theoretical basis and new therapeutic ideas for radiosensitization of hepatocellular carcinoma.Methods 1.The inhibitory effect of bufalin on the proliferation of human hepatoma HepG2 cells at different concentrations was determined by MTT assay.2.Cloning and formation experiments were conducted to analyze the combined effects of budialin and radiotherapy,and the cell survival curve and mean lethal dose(D0,quasi-threshold dose Dq,radiosensitization ratio SER,etc.)were obtained.3.Flow cytometry was used to analyze the effects of bufalin combined with radiation on cell cycle and apoptosis of human hepatoma HepG2 cells.Results 1.In toad venom determined by MTT method,the spirit of hepatocellular carcinoma HepG2 cells inhibit growth,compared with unused toad venom spirit of the control group,toad venom spirit of hepatocellular carcinoma HepG2 cells in a dose dependent inhibition rate gradually increased,namely with the increase of drug concentration,toad venom spirits of human hepatocellular carcinoma HepG2 cells inhibition rate increased,compared with the blank group,each group inhibition rate were increased(P < 0.05).The IC50 and IC20 of budialin against HepG2 cells were calculated as 1977.127nmol/L and 4.221nmol/L for 24 h.2.Cloned to form the experimental results show that after processing spirit toad venom D0 HepG2 liver cancer cells,Dq values were lower than simple radiotherapy group,radiotherapy sensitization than SER is 1.2,the difference statistically significant(P < 0.05),and prove that toad venom spirit enhanced radiation on human liver HepG2 cell killing effect,but also proves that under the same concentration of toad venom spirit,with the increase of irradiation dose of increased role for human hepatocellular carcinoma HepG2 cells.3.Approved by flow cytometry instrument after detected the toad venom spirit effect after 24 h,cells in the G2 / M ratio from 14.6% to 17.6%,compared with the radiotherapy,toad venom spirit + G2 / M phase proportion radiation group increased more obviously(P < 0.05),and proves the toad venom spirit can block cells more sensitive to ray G2 / M phase,namely the toad venom spirit improves the sensitivity of human hepatocellular carcinoma HepG2 cells to radiation,compared with the blank control group,each group can increase the rate of apoptosis of human hepatocellular carcinoma HepG2 cells,and toad venom spirit + radiotherapy group compared with the other groups effect is more significant(P < 0.05),The results showed that bufalin could increase the rate of cell apoptosis induced by radiation.Conclusions 1.Bufalin has inhibitory effect on the proliferation of human hepatocellular carcinoma HepG2 cells in vitro in a concentration-dependent manner.2.Bufalin can sensitize human hepatocellular carcinoma HepG2 cells by radiotherapy in vitro.3.The mechanism of bufalin increasing radiosensitivity of human hepatocellular carcinoma HepG2 cells cultured in vitro may be cell cycle arrest and apoptosis induction.
