| Objective: To investigate the effect of re Combinant adeno-associated virus Carrying tumor suppressor protein STAT1 in the treatment of glioma;to elaborate the Characteristic that the genetically modified adeno-asso Ciated virus can pass through the blood-brain barrier.After the tail vein injection of STAT1 gene into the mouse glioma in situ model,the targeted treatment of glioma is expected to play a therapeutic effect on tumor.Methods: 1.The STAT1 plasmid was constructed,identified and packaged with adeno-associated virus.2.GL261 cells were used to detect the transfection of adeno-associated virus.3.GL261 cells were used to establish the mouse glioma in situ model,and the constructed adeno-associated virus carrying STAT1 gene was injected into the tail vein to treat the mouse glioma in situ model,and the antitumor effect of STAT1 was observed in vivo.4.The establishment of tumor model in situ was evaluated according to the survival status of mice;the expression of STAT1 was detected by immunohistochemistry;the expression of STAT1 gene and protein was detected by RT-PCR and Western blot;the survival difference was analyzed acording to the survival time of mice.5.Western blot was used to detect some apoptosis related proteins,such as BCl-2,Bax,Caspase 8 and 9,to explore the possible mechanism.Results: STAT1 mediated by recombinant adeno-associated virus vector could in Crease the survival time of mice.Western blot and RT-PCR were used to detect the expression of STAT1 m RNA and protein in cells and tumor tissues of mouse model.The expression of BCl-2,Bax,Caspase8 and Caspase9 in tumor tissue was significantly increased by Western blot.Con Clusion: 1.The expression of STAT1 gene and protein increased after tail vein injection of adeno-associated virus carrying STAT1 gene into the mouse model of tumor.AAV can pass through the blood-brain barrier and promote the expression of STAT1 in tumor tissue.2.Recombinant adeno-associated virus has no obvious toxicity to heart,liver,spleen,lung and kidney.3.After injection of STAT1 gene carried by adeno-associated virus via tail vein,the targeted therapy effect on mouse glioma in situ model is obvious.Recombinant adeno-associated virus can increase the survival time of mice,and its mechanism may be related to apoptosis and necrosis pathway. |
