| Objectives: Through bioinformatics analysis,comparing the single-cell transcriptome sequencing expression profiles between human prostate mesenchymal stem cells(h PMSCs)and human prostate cancer cells(PCa),to investigate the transcriptome differential expression profiles of h PMSCs and PCa cells.Through bioinformatics to find out the genes related to PCA cell proliferation,invasion,apoptosis and drug resistance,to explore the genes involved in mesenchymal stem cells and tumor transformation Method: Four PCa cells were set as the experimental group(T group,including T1,T2,T3 and T4 groups);One case of h PMSCs was set as control group(group C).Collect the above cells,use Illumina sequencing platform for single-cell transcriptome sequencing.After sequencing quality control,quantify each gene expression level,FPKM was used as the expressed value.Then,the differential gene expression of each group was analyzed using DEseq with |Log2Fold Change| ≥1 and P <0.05 as the criteria,obtain differential gene expression profiles of each group,analyze the up-regulated and down-regulated genes.Finally,by comparing and screening the significantly different genes in each group,further analyse the differences of gene associated with PCa.Results: Through transcriptome high-throughput sequencing and bioinformatics analysis,the data of each group were compared and analyzed,the results were as following:(1)There were a total of 11,272 differentially expressed genes in T1_C,including 6080up-regulated genes and 5192 down-regulated genes.Among the top 10 genes up-regulated and down-regulated,CD74 and PTAFR were related to cell proliferation,LCP1,Sell and FGR were related to cell migration,ISG15,FITM3 and FKBP8 were related to cell immunity,LUM,DDX17 and CD63 were related to tumor suppression,and MALAT1,DCN and FPR1 were related to cell pathway.(2)There were a total of 11,915 differentially expressed genes in T2_C,including 6874up-regulated genes and 5041 down-regulated genes.Among the top 10 genes up-regulated and down-regulated,CD74,DDK1 and FGF7 were related to cell proliferation,LCP1,Sell,FGR and SPARC were related to cell migration,ISG15,FITM3 and FKBP8 were related to cell immunity,DDX17 and ANXA1 were related to tumor suppression,and MALAT1 and FPR1 were related to cell pathway.(3)There were a total of 11,216 differentially expressed genes in T3_C,including 4771up-regulated genes and 6445 down-regulated genes.Among the top 10 genes up-regulated and down-regulated,CD74 is related to cell proliferation,Sell,LCP1,FGR and HAS2 are related to cell migration,ISG15,FITM3 and FKBP8 are related to cell immunity,LOX is related to tumor suppression,and FPR1 and DCN are related to cell pathway.(4)There were a total of 12,005 differentially expressed genes in T4_C,including 8057up-regulated genes and 3948 down-regulated genes.Among the top 10 genes up-regulated and down-regulated,CD74 and TIMP1 are related to cell proliferation,LCP1,FGR,Sell and FN1 are related to cell migration,ISG15 and FITM3 are related to cell immunity,LUM,CD63 and LOX are related to tumor suppression,and FPR1,MALAT1 and DCN are related to cell pathway.(5)Then sort the significant difference genes from highest to lowest in the order of difference degree(|log2Fold Change|),the top 10 of difference degree of up-regulation and-down genes regulation associated with prostate cancer were summarized as follows:CD74,FPR1,MALAT1 and DCN may affect the proliferation of PCA cells by controlling the signal transduction pathway of PCA related cells.IFIM3,ARHGDIB,SPARC and CAV1 may affect the progression of PCA by regulating the EMT process.Lum,FN1 and LGALS1 may change the cell morphology by interacting with the surface of PCA cells and thus inhibit the progression of PCA.Conclusion:(1)In this study,single-cell transcriptome sequencing was performed on 4 groups of PCa cells and 1 group of h PMSCs using high-throughput sequencing technology.Through screening,sequencing and analysis of differentially expressed genes in each group,the top 10 significantly up-regulated genes were CD74,Sell,LCP1,ISG15,FGR,IFITM3,ARHGDIB,FPR1,TIMP1 and SPARC,while the top 10 significantly down-regulated genes were MALAT1,DCN,LUM,ANXA1,FN1,LGal S1,BCM,CD63,CAV1 and DDX17.(2)The significantly different genes associated with PCA were mainly related to the regulation of cell signal transduction pathway,EMT and the change of cell membrane structure. |
