Objective: This study used real-time quantitative reverse transcription polymerase chain reaction(RT-q PCR)to detect the expression of ATG7 and FIP200 in tongue squamous cell carcinoma and adjacent tissues,aiming to analyze the role of ATG7 and FIP200 in tongue squamous cell carcinoma and Clinical significance.On this basis,further experimental study and mechanism of autophagy inhibitor chloroquine on tongue squamous cell carcinoma were carried out to provide preliminary experimental basis for targeted therapy of tongue squamous cell carcinoma.Methods:Firstly,the expression of ATG7 and FIP200 m RNA in tongue squamous cell carcinoma and its adjacent tissues was detected by real-time quantitative reverse transcriptase polymerase chain reaction(RT-q PCR),and the correlation between the expression of ATG7 and FIP200 gene and clinicopathological factors of tongue squamous cell carcinoma was analyzed by statistical analysis of the correlation between the expression of ATG7 and FIP200 genes and clinicopathological factors of tongue squamous cell carcinoma by real-time quantitative reverse transcriptase polymerase chain reaction(RT-q PCR).In addition,the inhibitory effect of autophagy inhibitor chloroquine on tongue squamous cell carcinoma in vitro was studied.CCK8 cell proliferation test was used to detect the effect of chloroquine on the proliferation of tongue squamous cell carcinoma.Scratching test was used to detect the effect of chloroquine on the migration of tongue squamous cell carcinoma.RT-q PCR and Western blotting(WB)were used to detect the expression of ATG7 and FIP200 in tongue squamous cell carcinoma treated with chloroquine.Results: The detection of clinical tissue specimens of tongue squamous cell carcinoma showed that the expression levels of ATG7 and FIP200 m RNA in tongue squamous cell carcinoma tissues were higher than those in adjacent tissues,and they were not significantly related to clinicopathological factors.In the cell experiment in vitro,it was found that the proliferation and migration ability of tongue squamous cell carcinoma cells were significantly inhibited by chloroquine treatment,and the inhibitory effect was more obvious with the increase of chloroquine concentration.Low concentration chloroquine had no effect on the expression of ATG7 and FIP200 in tongue squamous cell carcinoma CAL-27 cell line.After high concentration chloroquine treatment,the expression of ATG7 was down-regulated,and the expression of FIP200 was not significantly changed.Conclusion:Compared with the adjacent tissues,ATG7 and FIP200 m RNA were highly expressed in tongue squamous cell carcinoma,indicating that ATG7 and FIP200 may be related to the occurrence and development of tongue squamous cell carcinoma.In the cell experiment in vitro,chloroquine could inhibit the proliferation and migration of tongue squamous cell carcinoma CAL-27 cells,and the inhibitory effect was more obvious with the increase of chloroquine concentration.High concentration of chloroquine can down-regulate the expression of ATG7;Chloroquine may play an inhibitory role in tongue squamous cell carcinoma,and it may be one of the effective strategies for the treatment of tongue squamous cell carcinoma.
