| Objective In this study,16s rRNA sequencing technology was used to detect the intestinal microecological characteristics of nude mice with gastric cancer treated with ACBP-OXA,and analyze the composition and diversity of intestinal microecological community of nude mice with gastric cancer treated with ACBP-OXA.To provide a new theoretical basis and reference for the treatment of gastric cancer with ACBP-OXA.Methods 1.Model establishment:human gastric cancer MKN-45 cells were cultured in vitro,and SPF male nude mice were selected as experimental animals,6-8 weeks of age.The inoculation site of gastric cancer MKN-45 cells was the right forelimb armpit and back of nude mice,and the inoculation concentration of gastric cancer MKN-45 cells was 2×10~6/m L.Each nude mouse was inoculated with 0.2m L.2.Forty nude mice successfully modeled were randomly divided into 4 groups with 10 mice in each group,which were Control group,ACBP group,OXA group and ACBP-OXA group,respectively,and intraperitoneally injected with drugs.The drug was given once daily for 14 days.3.Nude mouse feces sample collection:the feces of nude mice were extracted in a sterile environment to avoid the influence of external flora on the intestinal flora of nude mice.4.Tumor formation volume of nude mice was recorded.5.HE staining was performed on tumor tissues in each group.6.Fecal genome extraction.7.16s r RNA V3-V4 variable region amplification and sequencing.8.Bioinformatics analysis:generated OTU table and analyzed sequencing data quality control.9.Analysis of bacterial community structural diversity.Results1.ACBP-OXA combined therapy can significantly inhibit subcutaneous tumor formation in nude mice with gastric cancer.ACBP-OXA combined therapy improves the efficacy of OXA and reduces the dosage of OXA,thus reducing the toxicity of chemotherapeutic drugs,which may have the effect of reducing toxicity and increasing sensitization.2.HE results showed that the tumor cells in the ACBP-OXA group were reduced;the nuclei were pale,and the hyaline changes.3.The results of16s r RNA sequencing showed that there was a close correlation among the groups,and the sequencing diversity index,richness index and depth index met the requirements of the experiment.4.ACBP treatment group and ACBP-OXA treatment group can improve and restore the intestinal bacterial flora disturbance caused by tumor by changing the intestinal bacterial distribution or diversity and bacterial community structure of mice.5.ACBP,OXA,and ACBP-OXA can inhibit inflammation and tumor development by increasing the relative content of Lachnospiraceae NK4A136 and Bacteroides.6.OXA and ACBP-OXA therapy may have therapeutic effects by reducing the content of Alistipes.7.ACBP and ACBP-OXA treatment may reduce the inflammatory response by reducing the contents of Rikenellaceae RC9 gut and Staphylococcus,thus inhibiting the occurrence and development of tumors.8.ACBP may inhibit the growth of tumor cells by promoting the colonization of Lactobacillus,which increases the content of Lactobacillus.Conclusion 1.ACBP-OXA significantly inhibits tumor formation in nude mice,and combined treatment can reduce the dose of OXA and improve the efficacy of OXA,which may have the effect of reducing toxicity and increasing sensitization.2.ACBP treatment group and ACBP-OXA treatment group can improve and restore the intestinal bacterial flora disturbance caused by tumor by changing the intestinal bacterial distribution or diversity and bacterial community structure of mice.3.ACBP-OXA treatment can reduce the inflammatory response by increasing the relative contents of Lachnospiraceae NK4A136 and Bacteroides,and reducing the contents of Alistipes,Rikenellaceae and Staphylococcus,thus inhibiting the occurrence and development of tumors.4.The advantage of ACBP treatment is to increase the relative content of Lactobacillus and improve the occurrence and development of tumors. |
