| Objective By detecting the expression of MTDH in primary breast cancer and axillary lymph node metastasis,and analyzing the relationship between MTDH and clinicopathological characteristics of breast cancer,we speculated the role of MTDH in the biological process of breast cancer and its significance in targeted therapy of breast cancer.Methods1.A total of 110 breast cancer patients were collected from May 2019 to July 2020 in the breast cancer area a,Affiliated Hospital of Inner Mongolia Medical University.According to whether there is axillary lymph node metastasis,there are two groups,including 57 cases in metastasis group and 53 cases in non metastasis group.The expression and intensity of MTDH in 57 axillary lymph node metastasis were measured,and the expression of MTDH in the primary breast cancer of the two groups was statistically analyzed.The role of MTDH in invasion and metastasis of breast cancer was deduced,and whether the expression was targeted in breast cancer was analyzed It has certain application value in the treatment.2.The expression of MTDH was related to the age,month,stage and molecular type of breast cancer patients.It was estimated whether MTDH was related to the malignant course of breast cancer The degree of the disease is related to the prognosis.Results1.MTDH was mainly expressed in the perinuclear and cytoplasm of breast cancer cells,which was brown granules.In axillary lymph node metastasis,MTDH was mainly expressed in the nucleus.2.The positive expression rate of MTDH in 110 primary breast cancer was 82.7%.The expression of MTDH in the primary tissues of breast cancer in the two groups was analyzed by chi square test.The results showed that there were 32 cases of high expression of MTDH in the non metastasis group,accounting for 60.4%,45 cases in the metastasis group,accounting for 78.9%,the difference was statistically significant(P < 0.05).Furthermore,when the MTDH expression intensity was divided into(-,+,+ +,+ + +),the number of cases with strong positive(+ + +)in primary breast cancer cells in the metastasis group accounted for more than half of the total number of54.4%,and accounted for 68.9% of the high expression cases(+ +,+ + +).The number of strong positive cases in the primary focus samples of the non metastasis group accounted for only 24.5% of the total,accounting for 40.6% of the high expression cases.The weak positive rate in the metastasis group was 10.5%,the negative was 10.5%,the weak positive in the non metastasis group was 15.1%,and the negative was negative.It was 24.5%.The expression intensity was measured by chi square test,and P = 0.013,the results were statistically significant.3.The high expression rate of MTDH was 78.9% in primary tumor and 75% in metastatic tumor75.4%.Further analysis showed that there were 32 cases with high expression in primary and metastatic lesions,accounting for 56.1% of the total 57 cases,1 case(1.75%)with low expression in primary lesions and high expression in metastatic lesions,13 cases(22.8%)with high expression in primary lesions and low expression in metastatic lesions,and 11 cases(19.3%)with low expression in both.The expression of MTDH was positively correlated with lymph node metastasis(P <0.01).4.The expression of MTDH in 110 cases of primary breast cancer tissues was statistically analyzed.The results showed that the high expression of MTDH was significantly correlated with primary tumor size,number of axillary lymph node metastasis,clinical stage and Ki-67,P < 0.05,but not with age,menstruation,histological grade,pathological type,ER,PR,Her-2 expression,P > 0.05.5.In triple negative breast cancer,the high expression rate of MTDH was 75%,slightly higher than Luminal A(69.1%),B(67.9%)and Her-2 overexpression(73.3%),but the difference was not statistically significant(P > 0.05).Conclusions1.In primary breast cancer,the positive expression rate of MTDH with axillary lymph node metastasis was higher than that without metastasis,suggesting that MTDH may be involved in the occurrence,progression and recurrence of breast cancer.2.The positive expression rate of MTDH in axillary lymph node metastasis was similar to that in primary lymph node metastasis,and both were highly expressed.The expression of MTDH and primary lymph node metastasis was correlated,which may be a new target for the treatment of breast cancer metastasis.3.The expression of MTDH was correlated with tumor size,clinical stage,lymph node metastasis and Ki-67 expression,but not with age,menstruation,ER,PR and Her-2 expression.It suggests that MTDH may be involved in the proliferation and metastasis of breast cancer,or may be used as a diagnostic marker of breast cancer. |
