Objective: To investigate the polymorphisms of Toll-like receptor 2(TLR2)gene rs2289318C/G,rs3804100C/T and rs4696480A/T in Guangxi Zhuang population,and to compare their distribution differences with different regions and races.To investigate the relationship between the rs2289318C/G,rs3804100C/T and rs4696480A/T polymorphisms of Toll like receptor 2(TLR2)gene and the occurrence and development of sepsis,and to analyze the effect of rs2289318C/G,rs3804100C/T and rs4696480A/T polymorphisms of TLR2 gene on the expression of soluble toll like receptor 2(s TLR2)in plasma of patients with sepsis,In order to screen out the risk genotypes and alleles of TLR2 gene in patients with sepsis,and provide a new theoretical basis for the early diagnosis and prognosis of sepsis.Methods: The control group was selected 210 cases of healthy from the health examination center of Affiliated Hospital of Youjiang Medical University for Nationalities in June 2018 to July 2019.The sepsis group was selected 217 inpatients from the Department of Intensive Medicine of the Affiliated Hospital of Youjiang Medical University for Nationalities,the Department of Intensive Medicine of the people’s Hospital of Guangxi Zhuang Autonomous region,the Department of Intensive Medicine of the people’s Hospital of Baise City and the Department of Intensive Medicine of the Affiliated Hospital of Guangxi Medical University.Single base extension technology and DNA sequencing were used to detect the genotypes of rs2289318C/G,rs3804100C/T and rs4696480A/T of TLR2 gene in210 healthy persons of Guangxi Zhuang nationality receiving health examination.The genetic polymorphisms were analyzed and the results were compared with the genotypes of African Ancestry in Southwest US(ASW),the ancestry of African Caribbean in Barbados(ACB),Yoruba in Ibadan,Nigeria(YRI),American(AMR),Japanese in Tokyo,Japan(JPT),European(EUR),South Asian(SAS)and Han Chinese in Beijing,China(CHB)published by Thousand Human Genome Project Database.Single base extension PCR technique and DNA sequencing.Three single nucleotide polymorphisms(Single Nucleotide Polymorphisms,SNPs)were detected in TLR2 gene rs2289318C/G,rs3804100C/T and rs4696480A/T in 210 Guangxi Zhuang health check-ups(control group)and 217 sepsis patients(sepsis group)in Guangxi Distribution of polymorphism at SNPs loci,To determine the TLR2 gene SNPs locus genotype and allele distribution in Guangxi Zhuang population and sepsis patients;Plasma s TLR2 concentrations were detected by enzyme linked immunosorbent assay(Enzyme-linked immunosorbent assay,ELISA),To analyze the effect of TLR2 gene rs2289318C/G,rs3804100C/T and rs4696480A/T locus polymorphism on plasma s TLR2 expression in sepsis patients.Results: In the Guangxi Zhuang population,there were no significant differences in the genotype and allele frequencies of rs2289318C/G,rs3804100C/T and rs4696480A/T loci of TLR2 gene between different genders(P>0.05).There was a statistically significant difference between the rs2289318C/G allele in the Guangxi Zhuang population and that in SAS(P<0.05),but there were no statistically significant differences between the genotypes at the rs2289318C/G locus of TLR2 gene in the Guangxi Zhuang population and those in SAS(P>0.05).There were no statistically significant differences in the rs2289318C/G allele and its allele frequencies between the Guangxi Zhuang population and ACB,ASW,YRI,AMR,JPT,EUR and CHB(P>0.05).There were statistically significant differences in the genotypes at the rs3804100C/T locus of TLR2 gene and its allele frequencies between the Guangxi Zhuang population and ACB,ASW,YRI,AMR,EUR,SAS and CHB(P<0.05).However,there were no statistically significant differences in the genotypes at the rs3804100C/T locus of TLR2 gene and its allele frequencies between the Guangxi Zhuang population and JPT(P>0.05).There was a statistically significant difference between the rs4696480A/T genotypes and alleles in the Guangxi Zhuang population and that in ACB,ASW,YRI,AMR,EUR,SAS(P<0.05),but there were no statistically significant differences between the genotypes and alleles at the 4696480A/T locus of TLR2 gene in the Guangxi Zhuang population and those in CHB(P>0.05),there were a statistically significant differences between the genotypes at the rs4696480A/T locus of TLR2 gene in the Guangxi Zhuang population and those in JPT(P<0.05).However,there were no statistically significant differences in the alleles at the rs4696480A/T locus of TLR2 gene frequencies between the Guangxi Zhuang population and JPT(P>0.05).There were CC,CG,GG genotypes and C,G alleles in the TLR2 gene rs2289318C/G locus sepsis group,Their distribution frequency was 5.99%,34.56%,59.45%,23.27% and 76.73% respectively,The distribution frequency of control group was 3.81%,37.62%,58.57%,22.62% and 77.38% respectively.rs2289318C/G locus co-dominance model,invisibility model,dominant model and allele sepsis patients had no correlation with the risk of sepsis in the control group carrying the same co-dominance model,invisibility model,dominant model and allele,There was no statistical difference(OR=0.905;95%CI :0.606~1.352,P>0.05)。There were CC,CT,TT genotypes and C,T Calleles of rs3804100C/T in sepsis group,and Their distribution frequencies were 7.37%,39.63%,53.00%,27.19%,72.81%,There were 4.29%,38.10%,57.62%,23.33%,76.67% respectively in the control groups.There was no correlation between the risk of sepsis in patients with sepsis of TLR2rs3804100C/T locus co-dominance model,invisibility model,dominant model and allele and control group carrying the same co-dominance model,invisibility model,dominant model and allele.And their were not statistically significant(OR=1.131;95%CI : 0.760~1.683,P>0.05).There were AA,AT,TT genotypes and A,T alleles of rs4696480A/T locus in the sepsis group,and 33.33%,51.43%,15.24%,59.05%,40.95% in control groups.Patients with sepsis with AT genotypes at TLR2 gene rs4696480A/T loci were at low risk of sepsis than controls with the same genotype,The difference was statistically significant(OR=0.587;95%CI:0.387~0.892,P<0.05).Patients with sepsis with AT+TT genotypes in rs4696480A/T site dominant model had a low risk of sepsis compared with the control group with AT+TT genotypes(OR=0.630;95%CI:0.426~0.933,P<0.05).The plasma s TLR2 level in sepsis group was lower than that in control group,but there was no significant difference(P>0.05).Conclusion:(1)The polymorphisms of TLR2 gene rs2289318C/G,rs3804100C/T and rs4696480A/T in Guangxi Zhuang population are different from those in different ethnic groups.(2)The rs2289318C/G locus of peripheral blood TLR2 gene in sepsis patients was mostly GG genotype or G allele.Most of the rs3804100C/T mostly TT genotype or T allele,and the rs4696480A/T locus is mostly AA genotype or A allele,Most of the rs4696480A/T loci were AA or A alleles.(3)TLR2 gene rs4696480A/T locus polymorphism may be associated with redced risk of sepsis,while rs2289318C/G rs3804100C/T locus polymorphism is not associated with sepsis risk.
