Cervical cancer is the second malignant tumor among women’s tumors in China and even the world,and it seriously threatens women’s health and life safety.Platinum-based chemotherapy is an indispensable treatment method in the treatment of advanced cervical cancer.Cisplatin is currently the most commonly used platinum complex in clinical chemotherapy,but patients are prone to many side effects and even drug resistance.The baicalein and wogonin extracted from natural plants have a definite inhibitory effect on cervical cancer and have little toxic and side effects on normal cells.Based on this,this study explored the effect and mechanism of baicalein-wogonin complex combined with cisplatin in inducing apoptosis of cervical cancer cells.In this study,cervical cancer HeLa cells were cultured in vitro,and drug intervention was performed when HeLa cells grew to the logarithmic phase.The negative control group,cisplatin group,baicalein combined with cisplatin group,wogonin combined with cisplatin group,and baicalein-wogonin complex combined with cisplatin group were set up.The CCK-8 method was used to detect the effects of different drug groups on cell proliferation in terms of time and concentration;the colorimetric method was used to detect the changes in the LDH activity of HeLa cells after different drug groups,and the Caspase-3 fluorescence analysis was used to detect HeLa cells after different drug groups.Caspase-3 activity;Western Blot method was used to detect the expression level of Akt and mTOR protein and the expression level of corresponding phosphorylated protein.In this paper,taking the pathway of inducing cell apoptosis as a clue,study the effect and mechanism of baicalein,wogonin and their complexes combined with cisplatin in inducing HeLa cell apoptosis.This study shows that baicalein,wogonin and their complexes can all enhance the proliferation inhibition and apoptosis induction effects of cisplatin on cervical cancer HeLa cells,and they are time-dependent.Baicalein,wogonin and the complex combined with cisplatin may activate cell apoptosis by promoting cell damage.Its mechanism of action may be to induce HeLa cell apoptosis by inhibiting the activation of PI3K/Akt/mTOR signaling pathway,and can enhance the sensitivity of cisplatin to induce HeLa cell apoptosis.This study provides a certain experimental basis for the study of wogonin and the baicalein-wogonin complex enhancing the sensitivity of cisplatin to induce apoptosis in cervical cancer cells.
