| Objective:Observe the influence of expression in terms of Compound Qinglongyi Capsule acting on glomerular podocyte proteins nephrin and podocin in the Adriamycin Nephropathy(AN)rats.The experiment aims to explore the molecular mechanism of Compound Qinglongyi Capsules in reducing proteinuria in rats with nephropathy.Methods:72 clean-grade male SD rats were selected,12 were randomly selected as the blank group.According to the Bertani method,the remaining 60 animals were established the rat model of adriamycin nephropathy in rats by a one-time tail vein injection of adriamycin at the dose of 6.5mg/kg.The rats were randomly divided into 4 groups after successful modeling:compound Qinglongyi group(Qinglongyi group),tripterygium wilfordii polyglycosides group(tripterygium wilfordii group),benazepril hydrochloride tablets group(Lotingxin group),Model control group(model group).Each group was given intragastric administration in a predetermined method for 4 weeks.The data collection included testing the 24h-UPr of the rats at 1st,2nd,and 4th weekends after intragastric intervention.The rats were anesthetized at the end of 4 weeks.Relative data collection in this stage were including Scr,BUN,TC,ALB,ALT and AST.The expression of nephrin and podocin proteins in renal podocytes was detected by immunohistochemistry,the expression of nephrin mRNA and podocin mRNA in renal podocytes was detected by real-time PCR,and the pathological changes of the kidneys were observed by transmission electron microscope.Results:After 4-week treatment,the general state of Qinglongyi group,Tripterysium Glycosides group,Lotensin group were better than before.All24h-UPr were lower than the model group(P<0.05),and the effect of Qinglongyi group and Tripterygium Glycosides group were similar,significantly stronger than that of Lotensin group(P<0.05).Compared with model group,the value of TC wasall decreased,and the Qinglongyi group was the most significant(P<0.05).The value of ALB was all increased,and the Qinglongyi group was the most significant(P<0.05).The expressions of nephrin,podocin protein and mRNA in podocytes were higher than the model group(P<0.05),and the results of Qinglongyi group was similar to the Tripterygium Glycosides group,which is significantly higher than that of the Lotensin group(P<0.05).The structure of podocytes in the kidney tissues of rats in each group was improved under the electron microscope,and the degree of foot process disappearance and fusion of glomerular podocytes in Qinglongyi group and Tripterygium Glycosides group were much more significantly reduced.Conclusion:1.Compound Qinglongyi Capsules can improve the general state of rats with adriamycin nephropathy,reduce 24 hour urinary protein quantity,decrease the content of serum total cholesterol and increase the content of serum albumin.2.Compound Qinglongyi Capsule can reduce kidney pathological changes in rats with adriamycin nephropathy.3.Compound Qinglongyi Capsule may increase the expression of nephrin,podocin protein and mRNA in the renal tissue of rats with adriamycin nephropathy to repair kidney damage and reduce proteinuria. |
