Effect Of Huayu Tongyang Recipe On Serum Levels Of IL-1β And IFN-γ Of Ulcerative Colitis Rats

Objective:This research established the rat model of ulcerative colitis,to observes the effect of Huayu Tongyang Recipe on the expression of interleukin 1β(IL-1β)and interferon γ(IFN-γ)in the serum of model rats,and explores the mechanism of Huayu Tongyang Recipe for the treatment of ulcerative colitis.Furthermore,aims to provide relevant basis for practical clinical diagnosis and treatment.Method:32 Wistar rats were adaptively fed for 1 week,then 8 of them were randomly selected as the blank control group.And the remaining 24 rats were given gavage combined with free drinking of 5% dextran sodium sulfate solution to induce replication of the UC rat model.After the 7th day of modeling,4 rats of model group and 2 rats in the blank control group were randomly selected to be sacrificed.The UC modles were confirmed by comparing the mucosal ulcers and the pathological morphology of the colon between the dead rats.Then the 17 surviving model rats were randomly divided into model control group,mesalazine group and Huayu Tongyang group.The rats were treated with corresponding drug enema intervention.Rats in the Huayu Tongyang group were given 0.54g/ml Huayu Tongyang Recipe Water Decoction enema,0.036g/ml mesalazine enema in the mesalazine group,normal saline enema in the blank control group and model control group,all the enema remained 2ml each time,once a day.Rats in each group ate and drank normally during the continuous enema for 10 days.During the experiment,the disease activity index(DAI)was used to assess the general condition of the rats.After 10 days of enema,blood was taken from the femoral artery and each rats’ colon was dissected.The degree of morphological damage of the rat’s colonic mucosa was assessed according to the Colon Morphological Injury Index(CMDI).Then made the tissue sections,observe the pathological changes of colon tissue under light microscope and evaluate histological damage index(TDI)after HE staining;extracted serum from femoral artery blood by centrifugation,detected the expression levels of IL-1β and IFN-γ in serum of rats by using enzyme-linked immunosorbent assay(ELISA).Result:1.General conditions and scoring: There are no significant differences in the mental state,food intake,stool condition,and coat color of the rats in each group before modeling.After the intervention of model building,compared with the blank control group,the model rats had worse mental state,slow response,lazy action and poor appetite.Besides,the hematochezia symptom gradually became more serious.The rats’ hair was dry and dull.And the weight reduced significantly.Also the DAI score of model rats increased obviously.After treatment,the rats in the Huayu Tongyang group and the mesalazine group gradually recovered mental state,got the gradually increase in activities,diet and weight,improved the stool characteristics,and the hematochezia was significantly alleviated.Compared with the model group,the DAI scores of rats in the Huayu Tongyang group and the mesalazine group decreased(P<0.01),and there was no statistically effective difference between the two groups(P>0.05).2.The gross morphological damage of the colon: the surface of the colon in the blank group was smooth,without congestion and edema,or ulcer formation;the color of colon mucosa in the model group was dark,with obvious congestion,edema,thickened intestinal wall,scattered ulcer foci,and peripheral hyperemia or erosion.Compared with the blank control group,the CMDI score of the model group was significantly higher(P<0.01).Compared with the model group,The intestinal mucosal state of the rats in the Huayu Tongyang group and the mesalazine group improved after treatment,even if there was still mild congestion and edema,the ulcer tended to be repaired,and there was no obvious congestion and edema in the mucosa around the ulcer.Compared with the model group,the CMDI scores of the Huayu Tongyang group and the mesalazine group were all decreased(P<0.01),and there was no statistically effective difference between the two treatment groups(P>0.05).3.Histological damage: Under light microscope,the structure of the colon mucosa in the blank control group was intact,and the glands were arranged neatly.There was no inflammatory exudation,necrosis and shedding.In the model group,the structure of the mucosal propria glands was mostly damaged with inflammation Cell infiltration and distortion of crypt structure.Compared with the model group,the inflammatory cell infiltration was attenuated in Huayu Tongyang group and mesalazine group,and the degree of gland damage was relieved.Compared with the blank control group,the colonic TDI score of the model group was remarkably increased(P<0.01).Compared with the model group,the TDI scores of the Huayu Tongyang group and the mesalazine group were both decreased(P<0.01),and there was no statistically effective difference between the two groups(P> 0.05).4.Serum IL-1β and IFN-γ contents: The serum IL-1β and IFN-γ contents of UC model rats were higher than the blank control group(P<0.05).Compared with the model group,IL-1βand IFN-γ levels of rats serum in Huayu Tongyang group and mesalazine group were significantly decreased(P<0.01).There was no significant difference in IL-1β and IFN-γlevels between the Huayu Tongyang group and mesalazine group(P>0.05).Conclusion:1.The enema of Huayu Tongyang Recipe is effective in treating UC rats,which can improve the general state of rats,reduce inflammation in colon tissue,and reduce the scores of DAI,CMDI and TDI;2.Huayu Tongyang Recipe may inhibit the expression of pro-inflammatory cytokines IL-1βand IFN-γ,and control the development of inflammation.Furthermore,Huayu Tongyang Recipe can improve intestinal immune function,and promote the recovery of UC injury.
