Atherosclerosis(AS)is cardiovascular disease serious harm to human health,is a complex disease pathological process with the participation of many factors.AS is closely related to lipid metabolism,clinical trials,and the indicators show that lipid metabolism disorders can lead to the development of AS Ligustrum lucidum is riching for a variety of active ingredients and has a wide range of pharmacological effects,including the treatment of anti-osteoporosis,anti-tumor,anti-inflammatory,liver protection,lowering blood sugar and blood lipids and other pharmacological activities.Studies have shown that,Ligustrum lucidum can have normal cholesterol and heart rate by adjusting it,thereby improving blood lipid levels and cardiac function.In addition,Ligustrum lucidum can inhibit the formation of atherosclerotic plaques in rabbit aorta and reduce the area of AS plaques.In this experiment,based on phytochemical separation,thin layer chromatography,open chromatographic column,preparation liquid chromatography,high performance liquid chromatography,nuclear magnetic resonance spectroscopy,high resolution mass spectrometry,polarimeter,circular dichroism chromatograph and other methods,14 compounds were isolated and identified from the Ligustrum lucidum,among them,including phenylethanol glycosides(salidroside and acteoside),iridoid compounds[oleuropein,2"S-ethoxy-oleuropein,(8E)-nuzhenide,alas-hanidoid A,isonuzhenide,(8Z)-nuzhenide A,nuzhenide G13,iso-oleonuzhendie,1’’’-β-D-glucosylformoside,(2"R)-2"-methoxyoleuropein,specneuzhenid],flavonoid(apigein-7-O-β-D-glucopyranoside).In this experiment,50μg/mL ox-LDL was used to induce RAW 264.7 macrophages to establish a foaming model.And use the positive drug simvastatin and the 8 compounds obtained from the previous identification.Oil red O staining and intracellular total cholesterol determination showed that salidroside and acteoside have good anti-atherosclerotic activity.Since the previous research team has conducted a systematic study on the anti-atherosclerosis of salidroside,further research will be carried out on the anti-atherosclerosis in vivo of acteoside.Next,based on the AS animal model formed by high fat-induced Apo E-/-mice,using lipidomics technology to characterize the lipid metabolism profile of mouse liver tissue and combining pattern recognition analysis methods to find abnormally fluctuating lipid species related to AS,to evaluate the pharmacodynamic effect of acteoside in inhibiting the progression of AS and its regulating effect on abnormal lipid metabolites.Chemical indicators and pathological results indicate that the model of atherosclerosis is successful,and suggests that acteoside has a certain intervention effect on atherosclerosis.Based on LC/MS technology to identify lipid substituting components from the liver,screen 51 up-regulated lipids and 34 down-regulated lipids(M group vs C group),The abnormal increase or decrease of lipids in the model group has an indicative effect on atherosclerosis.Acteoside can simultaneously regulate33 lipids closely related to atherosclerosis,including 5 kinds of up-regulated TG lipids and 28kinds of down-regulated lipids including 8 types(TG,PC,LPC,CL,PE,SM).In the early stage of this article,14 chemical constituents were obtained by studying the chemical constituents of Ligustrum lucidum.Oil red O staining and determination of intracellular total cholesterol content were used to screen that salidroside and acteoside have good anti-atherosclerotic activity.Using high-fat diet to induce Apo E-/-mice to construct atherosclerosis model,preliminary verification that mullein can play an anti-atherosclerotic effect.
