As a special cultural carrier,wine occupies a unique position in human communication.Wine culture has penetrated into every field of human social life.Alcohol is not only a carrier of culture,but also a way for people to release pressure and emotions.Many people become addicted to alcohol,which leads to various diseases in the long run.With the development of society,the number of women drinking alcohol has been increasing year by year.A number of studies have shown that alcohol exposure during pregnancy can cause multi-dimensional damage to the embryo,and drinking by pregnant women increases the rate of abnormal embryo development,which has long-term adverse effects on both preimplantation embryos and post-birth fetuses.Therefore,how to repair the embryo damage caused by alcohol has become an important problem.However,traditional Chinese medicine is a multi-purpose compound decoction for fetal protection,and there are few studies on single active ingredient.This study intends to study the repair effect of single active ingredient "Liquiritin" on mouse embryos damaged by alcohol through the pharmacological method of traditional Chinese medicine.In this study,the method of in vitro fertilization was used to establish an in vitro culture system,adding alcohol to the medium,and exposing the pre-implantation embryos to 1.0% alcohol medium to analyze the direct effects of alcohol on the development of mouse embryos.100 μmol/L Liquiritin was added to investigate whether it could repair the embryo.The experiment was divided into five groups:control group,DMSO group,alcohol /DMSO group,alcohol/Liquiritin /DMSO group and Liquiritin /DMSO group.By observing each embryo developmental situation of each period,statistical blastocyst development rate,test important genes expression and proliferation in the process of embryonic development situation,research into alcohol before treatment and alcohol after adding licorice nucleoside under this condition,human preimplantation embryo development condition,after implantation embryo fetal placenta during the period of 13.5 days.In order to explore and analyze the repair effect of liquiritin on mouse embryos damaged by alcohol and the possibility of repairing placenta damage.The results showed that the alcohol exposure of preimplantation embryos led to slow embryo development,significantly reduced blastocyst development rate,significantly reduced embryonic cell number and proliferation rate,and the alcohol exposure of preimplantation embryos led to the destruction of the interaction between Cdx2 and Oct4 and the imbalance of expression,leading to the abnormal development process of late embryos.The expression of caspase-3 increased significantly.However,the addition of liquiritin after alcohol treatment can restore the rate of embryonic development,significantly increase the blastocyst development rate,and increase the number and proliferation of pre-implantation embryonic cells.At the same time,the abnormal imbalance of Cdx2 and Oct4 expression can be restored to a certain extent and can inhibit the apoptosis caused by alcohol to a certain extent.At the same time,alcohol exposure of preimplantation embryos will also lead to decreased implantation rate or non-implantation of embryos after implantation,reduced number of villi in the labyrinth region of placenta,smaller fetal blood vessels,maternal blood sinuses,lumen and blood volume.The addition of liquiritin after alcohol treatment can repair the quality of embryos,improve the abundance of villi in labyrinth region to some extent,and improve the reduction of fetal blood vessels,maternal blood sinuses,lumen and blood volume caused by alcohol.7.The addition of liquiritin after alcohol treatment can improve the abundance of villi in labyrinth region to a certain extent,and improve the reduction of fetal blood vessels,maternal blood sinuses,lumen and blood volume caused by alcohol.In conclusion,liquiritin plays an important role in repairing alcohol damage in embryos,including improving the development speed of pre-implantation embryos,improving the development rate of blastocyst,promoting the proliferation of embryonic cells,stabilizing the expression of important genes in embryonic development,improving the implantation rate after implantation,and improving the damage related to important blood vessels in placenta,etc.
