Biomarker protein immunoassay is a technology that is widely used and sensitive in medical diagnosis.With the continuous development of related science and technology,many researchers have proposed new technologies to replace traditional protein detection methods,magnetic probe immunochromatographic test paper Article technology is one of them.Aiming at the problems of high complexity and low portability of protein immunoassays for various existing markers,this thesis designs and develops a protein analysis system for magnetic probe-labeled immunochromatographic test strips.The main research contents of the thesis are as follows:(1)Introduce the development and research status of magnetic probe immunochromatographic protein detection technology,study the basic principles of magnetic probe quantitative analysis and immunochromatographic detection,and realize the first-level sensor that can be applied to the entire system,namely chromatography Test strip device,and then discuss the theoretical basis of magnetic probe protein analysis method applied to immunochromatography to achieve quantitative detection.(2)In response to the requirement of immunochromatographic protein detection,a magnetic probe suitable for this detection method is proposed.Subsequently,magnetic nanoparticles with good morphology,uniform dispersion and rapid magnetic response were realized,and then the prepared magnetic nanoparticles were functionalized into magnetic probes suitable for immunochromatographic protein detection through further modification.Magnetic nanoprobes can display the traces of marker proteins by using changes in their own magnetic properties,which is the key to the magnetoimmune protein analysis system.(3)Aiming at the characteristics of high reliability and portability of the proposed magnetic immune analysis system for field detection,a signal detection unit based on magnetoresistive sensing is proposed.The magnetic immunoassay instrument was realized on the basis of this.The instrument consists of two parts,namely the hardware part and the data processing and display part.The hardware part includes signal detection unit,mechanical transmission unit,power supply unit and central control unit.The data processing part is the key link of the whole system to realize the quantitative detection function.First,the possible conditions that affect the magnetic signal detection are studied,and then the magnetic signal is denoised,filtered and weak signal reconstruction operations,and finally the detection results are displayed in the display function.Unit.(4)In order to verify the practical application capabilities of the proposed magnetic immune protein analysis system,and to pinpoint the severe new coronary pneumonia epidemic currently facing the world.The proposed magnetic immunoassay instrument was successfully applied to the simultaneous quantitative detection of neocoronavirus antibody immunoglobulins M and G.Experimental data proves that the instrument can reach the sensitivity of 5 ng/m L and 10 ng/m L for two proteins within 15 minutes,and it has the ability to be applied to actual medical field detection in the future.(5)Summarized and thought about the research content of the full text,and then looked forward to the future work content.
