Objective:An in vitro study on the anti-phlogosis function of medician based on the MAPK signaling pathway was conducted to clarify the anti-inflammatory effect of 6-gingerol.Method:1.6-Gingerol is purchased from a biotechnology company,and the purity of > is required to be 98%.Before use,the medium is prepared into a storage solution with a concentration of 250 mm and stored at-20℃.Dilute to the required experimental concentration before the experiment,and use after filtration.2.MTT method and LDH method were used to determine the concentration of drug safe action.3.The level of NO release decreased with the increase of drug concentration.4.The secretion of inflammatory cytokines,including IL-6 and IL-10,was detected by ELISA kit.5.RT-qPCR detection and related factor expression level in cells,including IL-6,IL-10,iNOS and cox-2,TNF,IL-1?.6.Gingerol decreased the protein phosphorylation levels of P38,ERK and JNK after LPS induction to different degrees.Result:1.MTT method was used to detect the effect of drugs on cell proliferation,and LDH method was used to detect the degree of cell damage.Through mutual verification,the effective concentration of 6-gingerol was determined to be below 20μmol/L.2.The effective concentration of gingerol inhibited the release level of NO in LPSinduced RAW264.7 cell inflammation model to different degrees(P<0.05)in a concentration-dependent manner.3.6-Gingerol inhibited the secretion of inflammatory cytokine IL-6 to a certain extent(P<0.05);4.6-Gingerol significantly decreased the m RNA expressions of IL-6,IL-10,iNOS and COX-2(P<0.05);5.6-Gingerol down-regulated the protein phosphorylation levels of P38,ERK and JNK after LPS-induction to varying degrees(P<0.05);Conclusion:6-gingerol May play an anti-inflammatory role by reducing NO production and IL-6 secretion and inhibiting the phosphorylation of key proteins P38,ERK1/2 and JNK1/2 in the MAPK pathway.
