| 1.Purpose1.1 Evaluation of the relative specific regulation of acupuncture at the five points of the heart and lung meridian on bronchial asthma in mice.1.2 To study the role of muscarinic receptors M1,M2,and M3 in bronchial asthma,and to explore the relative specific mechanism of action between Taiyuan and the lungs.2.Method2.1 Using hematoxylin-eosin staining lung tissue method to evaluate the effect of acupuncture at the five points of the heart and lung meridian on bronchial asthma2.2 Select 130 healthy adult female BALB/c mice.After 1 week of adaptive feeding,the animals are randomly divided into normal group,model group,acupuncture at Shaoshang acupoint group,acupuncture at Yuji acupoint group,and acupuncture at Taiyuan acupoint group,Acupuncture Jingqu points group,acupuncture Chize point group,acupuncture Shaochong point group,acupuncture Shaofu point group,acupuncture Shenmen point group,acupuncture Lingdao point group,acupuncture Shaohai point group.9 in each group.Acupuncture at the five Shu points of the Heart and Lung Meridian selects bilateral acupoints for one week of intervention.The changes in airway resistance and lung airway compliance of mouse breathing air measured by Ani Res2005 animal lung function analysis system.2.3 Twenty-four adult and healthy female BALB/c mice were selected.After 1 week of adaptive feeding,the mice were randomly divided into normal control group,asthma model group,and acupuncture Taiyuan acupoint group,with 6 mice in each group.Acupuncture selects Taiyuan points on both sides.Intervention treatment for one week.Mice lung tissues were collected after treatment.Real-time fluorescent quantitative PCR(RT-PCR)method was used to detect the changes of muscarinic receptor M1,M2,M3 m RNA content in the lung tissue of each group of mice.Western blotting(WB)was used to detect and compare the expression of muscarinic receptor M1,M2,and M3 protein in the lung tissue of each group of mice.3.Results.3.Results3.1 The pathological changes of mouse lung tissues were observed by hematoxylin-eosin staining.The results showed that the lung pathological damage of mice was reduced after acupuncture at five points of the heart and lung meridian,and the five points of lung meridian was better than five points of heart meridian.3.2 The Ani Res2005 animal lung function analysis system was used to detect and compare the airway resistance and lung airway compliance of each group of mice in exhalation and inhalation.The results showed that for the inhalation and expiration airway resistance model group,the airway resistance increased significantly,and the inhalation and expiration airway resistance decreased to varying degrees after the intervention of different acupuncture points.Acupuncture at the five points of the lung meridian was better than acupuncture at the five points of the heart meridian.The decline is more pronounced.For the lung airway compliance model group,the lung airway compliance decreased significantly,and the lung airway compliance increased to varying degrees after the intervention of different acupuncture points.Acupuncture at the five points of the lung meridian increased more significantly than acupuncture at the five points of the heart meridian.3.3 Western blot was used to detect and compare the protein expressions of M1,M2 and M3 receptors in lung tissues.The results showed that the protein expressions of M1 and M3 receptors were significantly increased and the protein expression of M2 was significantly decreased in mice after modeling.After acupuncture at Taiyuan point,the protein expressions of M1 and M3 receptors were both decreased,while the protein expression of M2 was increased.Real-time quantitative PCR(RT-PCR)was used to detect and compare the changes of M1,M2 and M3 receptor m RNA contents in the lung tissues of mice in each group.The results showed that the expressions of M1 and M3 m RNA were significantly increased and the expression of M2 m RNA was decreased after the modeling of mice.After the intervention of acupuncture at Taiyuan point,the expressions of M1 and M3 m RNA were decreased and the expression of M2 m RNA was increased.4 Conclusion4.1 Acupuncture at the five points of the lung meridian has a better effect than acupuncture at the five points of the heart meridian in the treatment of bronchial asthma.Taiyuan point has a significant effect in the five points of the lung meridian,which confirms the related theories of meridians and viscera.4.2 Acupuncture at the Taiyuan point of the lung meridian can treat asthma,possibly by adjusting the expression of cholinergic receptors M1,M2,and M3. |
