| [Objective]:To use fecal microbiota transplantation(FMT)to treat common ulcerative colitis(CUCM)and damp ulcerative colitis((DUCM)in mice to further detect various indexes of mice to verify the efficacy of FMT and the bitter cold taste of FMT(New Jinzhi).[Methods]: The mouse CUCM model was established by DSS induction + normal diet + normal temperature feeding,and the DUCM model was established by DSS induction + high-fat and high-sugar feeding + hot and humid environment.After successful establishment of the model,FMT and 5-aminosalicylic acid(5-ASA)were given respectively.The changes of intestinal tissue were observed by HE staining,the ultrastructural changes of tissue were observed by transmission electron microscope,the contents of red blood cells,white blood cells and platelets in blood were detected by blood routine,flow cytometry and high-throughput 16 s sequencing were used to detect the distribution of microflora in intestinal contents.[Results]: HE staining of intestinal tissue showed that there were varying degrees of defect or exfoliation and necrosis on the surface of intestinal mucosa in CUCM and DUCM model groups,partial formation of ulcers,disappearance of crypt,infiltration of a large number of inflammatory cells in lamina propria,thickening and edema of mucosal muscle layer,vasodilation of submucosa,and infiltration of more red blood cells and eosinophils.It was significantly improved after FMT treatment,and the glands of the DUCM+FMT group were neatly arranged and compactly arranged,and the incomplete fractures were less than those of the CUCM+FMT group,compared with the CUCM+FMT group.The ultrastructure of intestinal tissue under transmission electron microscope showed that the surface microvilli of epithelial cells in CUCM and DUCM model groups were sparse,different in length,irregular in shape,widening of cell junction space,decrease of goblet cells,vacuoles in cytoplasm,swelling of mitochondria,disappearance of some cristae,expansion of endoplasmic reticulum or vacuolar changes in some cases,which were significantly improved after FMT treatment,and the microvilli of DUCM and CUCM model groups were tighter than those of the latter after FMT treatment.There are also more goblet cells than the latter.Flow cytometry showed that Th1 cells increased significantly and Th2 cells decreased significantly in CUCM group and DUCM group.After treatment with FMT and 5-ASA,there were significant improvements in DUCM model group,Th1 cells decreased significantly,Th2 cells significantly increased,close to the normal level,while in CUCM model group,Th1 cells increased slightly and Th2 cells decreased slightly after FMT treatment,while Th1 cells decreased and Th2 cells increased after 5-ASA treatment.Blood routine test showed that the number of leukocyte(WBC)and platelet(PLT)in CUCM and DUCM model group increased significantly,while erythrocyte(RBC)and hemoglobin(HGB)decreased.After FMT treatment,the blood routine data were improved,and WBC,RBC,PLT,HGB was close to the normal control group.The decrease index of leukocyte(WBC)and platelet(PLT)and the increase index of erythrocyte(RBC)and hemoglobin(HGB)in DUCM+FMT group were better than those in CUCM+FMT group.According to the curative effect index analysis,the scores of disease activity index of mice in each group after treatment were lower than those before treatment,CUCM+FMT and CUCM+5-ASA,DUCM+FMT and DUCM+5-ASA,P > 0.05,indicating that there was no significant difference between the two groups,which proved that the two groups were effective.The curative effect index of DUCM+FMT model group was better than that of CUCM+FMT model group.Flora analysis showed that in CUCM and DUCM model groups,the abundance of Ruminococcus decreased significantly,while that of Akkermansia increased significantly.After FMT treatment,there is a tendency to gradually close to the normal group,indicating that the intestinal flora is improving.[Conclusion]: FMT has a good effect on UC model mice,and the effect on DUCM type mice is better,which proves that New Jinzhi has bitter cold taste.It is speculated that its effect may be through the regulation of intestinal flora to improve the balance of Th1/Th2 cells to achieve the purpose of treatment. |
