| Research objectives:By observing the effects of Shenqi Jianxin recipe on cardiac function and calcium regulatory protein in rats with chronic heart failure,the effectiveness and possible mechanism of Shenqi Jianxin recipe in the treatment of chronic heart failure were further clarified.Research method :80 male SD rats of grade SPF were selected,Weight 220±250 g,The adaptive feeding group was randomly divided into sham operation group(n=10)and model group(n=70),The rat model of chronic heart failure after myocardial infarction was established by left coronary artery ligation,Among them,The sham operation group used the same operation method but only threading without ligation.Four weeks after surgery,The surviving rats were randomly divided into model group,benazepril group,Shenqi Jianxin prescription high dose group,middle dose group and low dose group.Benazepril suspension 1.2 g/kg/d gavage;Traditional Chinese medicine high dose group 14.8 g/kg·d、 medium dose 7.4 g/kg·d、 Chinese medicine low dose group 2.7 g/kg·d gavage,And the model group and sham operation group were given 3ml·d saline,Drug intervention lasted 4 weeks.After four weeks of continuous intervention,Color Doppler echocardiography was performed in all rats to assess cardiac structure and cardiac function,left ventricular ejection fraction(LVEF),left ventricular short axis shortening rate(LVFS),left ventricular end-systolic diameter(LVIDs),left ventricular end-diastolic diameter(LVIDd),left ventricular end-systolic volume(LVESV),left ventricular end-diastolic volume(LVEDV).Death to rats,Blood samples were collected by abdominal aortic blood collection,The BNP、AngⅡ、ALD、ET-1 content in serum was detected by ELISA detection method.The heart tissue was cut off immediately after death and the left ventricle was isolated,Rinse with ice saline and dry with filter paper,weigh separately,Cardiac weight(HW),Total weight/weight= HW/BW,calculated heart weight/body weight).Split the broken ventricular tissue into two parts,A portion is fixed in 4% paraformaldehyde solution and stored in 4℃refrigerator,Preparation for pathological sections HE staining and Masson staining for cardiomyocyte morphology and myocardial fibrosis observation;The other part was stored in the-80 ℃ refrigerator for Western Blot detection and m RNA content in myocardial tissue.Findings:(1)General observation: false surgery group in good mental state,free activity,sensitive reaction,hair color normal luster;model group: fatigue,reduced activity,depressed,depressed,crouching,dull reaction,yellow luster,easy hair,some rat diarrhea,limb edema,abdominal bulge,and purple dark tongue.Ddose of Shenqi Jianxin: good mental state,general activity,hair underluster,normal diet and drinking water,no diarrhea,no obvious limb edema.Benapril group: slightly slow reaction,good activity,general mental state,and undershiny hair.(2)Echocardiographic results: compared with the model group,LVIDs,LVIDd,LVESV,LVEDV was significantly lower and significant(P <0.01);compared with the model group,the middle,high,low dose and Benappli group could increase LVEF,LVFS,and LVIDs,LVIDd,LVESV,LVEDV decreased and difference was of statistical significance(P <0.05).(3)Serum BNP,AngⅡ,ALD,ET-1 content: compared with the false surgery group,the serum content was significantly higher and significant(P <0.01)and the serum decreased in all dose and Benaprii groups compared with the model group(P <0.05).(4)SERCA2a,Ry R2,Ca MKⅡ,NCX protein content and m RNA level of myocardial tissue: compared with the false surgical group,Ca MKⅡ,NCX protein and m RNA in myocardial tissue in the model group,SERCA2 a,Ry R2 protein content and m RNA,Statistical significance of the difference(P <0.01);In comparison to the model groups,Ca MKⅡ,NCX content in all dose and Benapril group,The SERCA2 a,Ry R2 protein content has increased significantly,The differences are statistically significant(P<0.05).(5)Heart weight index: compared with the false surgery group,rat weight loss,cardiac weight increase,cardiac weight index was significantly higher than the prosthetic surgery group(P <0.05),the cardiac weight index of SQ and Benapril group had statistical significance(P <0.05).(6)HE staining of myocardial tissue: regular structure,normal arrangement,normal morphology,no hypertrophic hypertrophy,no inflammatory cell infiltration in the model group,irregular size,irregular arrangement,uneven staining of cardiac fibers,obvious interstitial hyperplasia,accompanied by inflammatory cell infiltration.The myocardial histopathy in the remaining drug intervention groups was significantly improved compared with the model group,the neat arrangement of myocardial cells,and the inflammatory response was relieved.Masson staining: No abnormal blue stained collagen fibers was found in the myocardial stroma in the pseudosurgical group.The large distribution of blue myocardial collagen fibers was seen in the model group,showing obvious myocardial fibrosis compared with the false surgery group;the myocardial fiber distribution area in the other drug intervention group decreased,and the degree of myocardial fibrosis was significantly improved compared with the model group.Conclusion:1.Shenqi Jianxin Decoction can reduce the heart weight index of rats with chronic heart failure,improve myocardial tissue lesions,inhibit myocardial fibrosis,delay myocardial remodeling and improve cardiac function.2.Shenqi Jianxin Decoction can reduce Ang II and ALD levels in rats with chronic heart failure,indicating that Shenqi Jianxin Decoction can inhibit the activation of RAAS system,enhance myocardial contractility and improve cardiac function.3.Shenqi Jianxin Decoction can reduce the levels of BNP and ET-1 in rats with chronic heart failure,regulate vascular endothelial function and protect the heart.4.Shenqi Jianxin Decoction can reduce the protein levels of Ca MKII,NCX and m RNA expression levels of Ca MKII,NCX in rats with chronic heart failure,increase the protein levels of SERCA2 a,Ry R2 and m RNA expression levels of SERCA2 a,Ry R2,maintain calcium homeostasis in myocardial cells,inhibit calcium overload,and improve cardiac systolic and diastolic function in rats with heart failure. |
