Objective:To preliminarily investigate the correlation of p21 and REST genes with neuroendocrine differentiation induced by mast cells in androgen independent prostate cancer cells.Methods:The aperture of 0.4μm Transwell chambers were used to tie human prostate cancer cells(PC-3)and mast cell co-culture,observed the PC-3 cell morphological changes with inverted microscope,tested the activity and proliferation of PC-3 cells by CCK-8 experiment and tablet clone formation experiment,and used q RT-PCR experiment to detect the markers of neuroendocrine(NSE,SYP,CHGA),p21 and REST m RNA level.Independent-sample t test was used for statistical analysis,and P<0.05 indicated statistically significant difference.Results:After co-culture with P815 cells,the morphology of PC-3 cells changed from spindle to longer,irregular shape with dendritic structure,and the intercellular space became larger.CCK-8 results showed that OD value of PC-3+P815 group was lower than that of PC-3group(P<0.05),but with the extension of co-culture time,OD value of PC-3+P815 group first decreased and then increased,and OD value was the lowest when P815 concentration was 10~4 per well.The results of tablet clone formation experiment showed that the cell clone formation rate of PC-3+P815 group with P815 concentration of 10~4 per well and 4 days of co-culture was lower than that in PC-3 group(P<0.05).The q RT-PCR results showed that compared with PC-3 group cells,the m RNA expression of NSE,SYP,and CHGA were significantly up-regulated(P<0.05),the m RNA expression of p21 was up-regulated(P<0.05),and the expression m RNA of REST was down-regulated(P<0.05)in PC-3+P815 group cells.Conclusion:Mast cells may induce neuroendocrine differentiation in androgen independent prostate cancer cells,in addition,p21 and REST genes are related to this process to some extent.
