| Objective:Systematic evaluation of the correlation between the expression level ofα-synuclein in cerebrospinal fluidand Alzheimer’s Disease provides a new direction for the diagnosis of AD and the study of its mechanism.Methods:Pub Med,Embase,CNKI,CBM,Wanfang and other databases were retrieved by computer,and all literatures from the establishment of the database to October 2020 were searched.Studies were screened basised on the inclusion and exclusion criteria,and the research data on the correlation between CSFα-synuclein and Alzheimer’s disease was extracted.The Newcastle-Ottawa Quality Assessment Scale(NOS)was adopted to assessthe quality of the collected literatures,and the low-quality literatures with a total NOS score of<5 were excluded to determine the final inclusion study.Rev Man5.3software was used for data pooled and analyzed.In addition,sensitivity and publication bias of the study were examined.P<0.05 was thought as statistical difference.Results:9 articles were included in all,including 685 cases in the patient group and 510 cases in the matched group.The results of meta-analysis showed that the concentration ofα-synuclein in the cerebrospinal fluid was significantly higher in the AD group than in the healthy/neurol control group(overall effect test Z=5.26,P<0.00001,the combined effect value was 0.53,95%CI=0.34~0.73,located on the right side of the invalid vertical line),with moderate heterogeneity among researches(P=0.009,I~2=59%).In subgroup analysis,the overall effect test of ELISA group(Z=3.45,P=0.0006)SMD was(0.52,95%CI=0.22~0.81).Overall effect test of Luminex group(Z=5.10,P<0.00001)SMD was(0.57,95%CI=0.35~0.78),indicating low heterogeneity.The overall effect test of healthy control group(Z=3.41,P=0.0006,)SMD was(0.32,95%CI=0.14~0.50),and the overall effect test of neural control group(Z=9.45,P<0.00001)SMD was(0.80,95%CI=0.63~0.97).The analysis heterogeneity of the two groups was low.Sensitivity analysis indicated that the results were credible.Conclusion:The concentration ofα-synuclein in cerebrospinal fluid was significantly higher than that in the control group,andα-synuclein in CSF is a potential biological marker for the diagnosis of AD. |
