Objective:To explore the expression mechanism and expression level of TRIM11 in colorectal cancer.Methods:1.RT-PCR analysis of TRIM11 m RNA expression in 69 pairs of colorectal cancer tissues.Then,Kaplan-Meier analysis of the relationship between TRIM11 expression and patient survival.2.By the Western blot,we analyze the protein expression levels of TRIM11 and H3K27 ac in 12 pairs of adjacent normal tissues and 12 pairs of colorectal cancer tissues.3.By transfecting TRIM11-Pex3 into SW620 and SW480 cells,it can construct overexpression cells.TRIM11-sg R was transfected into DLD1 and Wi Dr cells to construct knockdown cells.4.By CCK8,cell colony formation experiment and Edu,this can detect cell proliferation ability in vitro.5.By Treating DLD1 cells with C646 and treating SW480 cells with a mixture of deacetylase inhibitors,then it can analyze the relations between the expression level of H3K27 ac and the expression level of TRIM11.Results:1.TRIM11 is highly expressed in colorectal cancer tissues,and it is related to the overall survival rate of colorectal cancer patients.2.In colorectal cancer tissues,H3K27 ac is highly expressed.Moreover,the expression of TRIM11 is significantly correlated with H3K27 ac.3.The datum of extracorporeal experiments show that TRIM11 can promote the proliferation of colorectal cancer cells.4.By treating cells with C646,the expression of TRIM11 increases with the increase of the degree of H3K27 ac.Then,by treating cells with the acetylase inhibitor mixture,the expression of TRIM11 decreases with the decrease of the degree of H3K27 ac.Conclusions:In colon cancer cells,TRIM11 is high expression and it promotes the proliferation of colon cancer cells.In addition,the high expression of TRIM11 may be related to the enrichment of H3K27 ac.
