Objective:To study the expression of USP13 in human breast cancer cell line MCF-7 and the effect of miR-130b-3p targeting USP13 on the function of breast cancer cells.Methods:The expression level of USP13 in human normal breast epithelial cells and human breast cancer cell line(MCF-7)was detected by real-time quantitative PCR;Ever build USP13 expression vector transfection breast cancer cell line MCF-7,and then use cell scratch experiment,Transwell cell migration and invasion detection after USP13 express MCF-7 cell proliferation,migration and changes of the invasive ability.Bioinformatics predicted and constructed miRNAmimic and its target gene wild type and mutant co-transfection cell lines,carried out double luciferase reporter gene experiment,detected the luciferase activity in the cells,and judged whether miRNA could target its target genes through specific binding sites.Results:1.The expression level of USP13 is relatively low in breast cancer cells(MCF-7)compared to normal breast epithelial cells(P < 0.01).2.Overexpression of USP13 could significantly inhibit the migration and invasion of MCF-7 cells(P < 0.001).3.There is a binding site of miR-130b-3p in USP13 sequence,and USP13 is the target gene of miR-130b-3p.Conclusion:1.USP13 is the target gene of miR-130b-3p.2.In breast cancer cells,overexpression of USP13 can significantly inhibit the migration and invasion of MCF-7 cells.
