Objective: To study the effect of gastrodin on the recovery of motor function and the expression of GAP-43 in animals with spinal cord injury.Methods : Thirty New Zealand white rabbits were randomly divided into sham operation group,control group,and experimental group,with 10 rabbits in each group.The sham operation group received only surgical exposure.The control group and the experimental group applied the modified Allen’s blow method to establish a spinal cord injury model.After modeling,the experimental group was given intravenous injection of 100 mg/(kg · d)of gastrodin,and the sham operation group and the control group were given 10 m L of normal saline intravenously for 7 consecutive days.1,3,5,and 7 days after modeling,the rats in each group were scored for neurological function;7 days after modeling,the injured spinal cord tissue was taken and stained with hematoxylin-eosin and Nissl,Western blot detection,immunofluorescence observation.The experimental protocol was approved by the Animal Experiment Ethics Committee of Guilin Medical College(GMC201805018).Results:(1)In the neurological function score,the sham operation group had normal function.1day after modeling,there was no significant difference between the experimental group and the control group.On 3,5,and 7 days after modeling,the hind limb score of the experimental group was significantly higher than that of the control group(P <0.05);(2)Hematoxylin-eosin staining showed that syringomyelia was seen in the control group,the structure was incomplete,the boundary between gray matter and white matter was unclear,and neurons in the gray matter were necrotic and apoptosis;the experimental group showed cavitation and necrosis,the integrity of spinal cord tissue was good,and gray matter nodes Relatively clear;(3)Nissl staining showed that there were no neurons in the control group,and a large number of glial cell proliferation was seen;in the experimental group,neurons survived,and glial cell proliferation was relatively less;(4)Western blot detection and immunofluorescence observation showed that the GAP-43 protein expression of the control group and the experimental group was higher than that of the sham operation group,and the experimental group was significantly higher than the control group.Conclusions:The results show that gastrodin can promote the recovery of motor function in spinal cord injury,and may promote the regeneration of spinal cord axons by increasing the expression of GAP-43 protein.
