Objective To study the expression of WNT inhibitory factor 1(WIF1)in clinical tissue samples of non-small cell lung cancer(NSCLC),analyze the relationship between the expression of WIF1 and the metastasis of NSCLC,study the fuctions of WIF1 on the invasion and migration of A549 cell,and explore the molecular mechanism of WIF1’s functions.Methods Clinical tissue samples of non-small cell lung cancer were collected(cancerous tissue and para-cancer tissue).Real-time PCR and Western blot were used to detect the expression of WIF1 at m RNA and protein levels in clinical samples,respectively.Meanwhile,bioinformatics analysis was performed on the data from the TCGA database to help verify the clinical test results.The relationship between WIF1 m RNA expression and clinicopathology and prognosis was analyzed.Lung cancer cells A549 were cultured,and WIF1 overexpressed plasmid and no-load plasmid were transfected with A549,respectively.The effects of WIF1 overexpression on cell invasion and migration were detected by scratch and Transwell experiments,and the expression levels of MMP-2 and MMP-9 were detected by Western blot.The expression of β-catenin was detected by Western blot to verify the effect of WIF1 on WNT /β-catenin signaling pathway.Results The expression of WIF1 m RNA and protein in the cancer tissues was significantly lower than that in the adjacent tissues(P < 0.05).Meanwhile,data from TCGA database showed that WIF1 expression abundance in lung adenocarcinoma and lung squamous cell carcinoma was significantly lower than that in normal lung tissue(P< 0.05).The incidence of lymph node metastasis and TNM stage of lung cancer patients with high WIF1 expression were significantly lower than those with low WIFI expression,and their survival was significantly higher than that of the low WIFI expression group(P < 0.05).Overexpression of WIF1 significantly inhibited migration and invasion of A549 cells,and significantly down-regulated the expression of tumor metastasis related markers MMP-2 and MMP-9(P < 0.05).Overexpression of WIF1 significantly inhibited β-catenin and Myc protein expression(P < 0.05).Conclusion The high expression of WIF1 can inhibit the invasion and migration of NSCLC cells by blocking the Wnt/β-catenin signaling pathway.
