Objective:Clarify the chemical components of Tuoli Xiaodu powder,use network pharmacology to predict its active ingredients,targets and pathways,and clarify its quality markers;establish a content determination method for multiple index components of Tuoli Xiaodu powder,and establish its fingerprint analysis method And then build the quality control standards of Tuoli Xiaodu powder.Methods:UPLC-Q-TOF/MS technology is used to analyze the full components of Tuoli Xiaodu powder to fully understand the structure of its compounds;apply network pharmacology methods to screen the active ingredients of Tuoli Xiaodu powder through the TCMSP database,and the PharmMapper database to screen active ingredients related targets,GeneCards、DrugBank、DisGeNet,OMIM database to screen disease-related targets,the intersection of the two targets is imported into Cytoscape software to construct a drug-ingredient-potential target network diagram,and then imported into Metascape platform for GO biological process enrichment analysis and KEGG metabolic pathway enrichment analysis;according to the selected potential quality markers,the HPLC multi-index component content determination and UPLC fingerprint of Tuoli Xiaodu powder were established.Results:(1)chemical composition analysis of Tuoli Xiaodu powder:a total of 10 8 compounds were analyzed and characterized,including 3 2 flavonoids,1 3 terpenoids,12 coumarins,1 3 saponins,9 alkaloids,10 phenol s,3 quinones,2 amino acids and 14 other components.According to the classification of medicinal materials,6 are from ginseng,4 are from Platycodon grandiflorum,32 are from licorice,3 are from chuanxiong,4 are from Paeoniae Alba,5 are from Angelica dahurica,5 are from Atractylodes macrocephala,9 are from Astragalus,9 are from angelica,12 are from honey suckle,2 are from Gleditsia sinensis,and 17 are multi drug.(2)Study on quality markers of Tuoli Xiaodu powder:27 ingredients were screened from the chemical composition database of Tuoli Xiaodu powder,and 78 potential targets were obtained by taking the intersection of ingredient targets and disease targets,constructing drugs-ingredients-potentials target network diagram,and the GO biological process enrichment analysis and KEGG metabolic pathway enrichment analysis of potential targets at the same time,predict that Tuoli Xiaodu powder will play a therapeutic effect through multiple components,multiple targets,and multiple pathways.With reference to 10 8 compounds obtained by full-component analysis and 27 predicted active ingredients,4 quality markers were screened for quality standard research.(3)Study on the quality standard of Tuoli Xiaodu powder:Establish a content determination method for the selected four quality markers.The quality markers included paeoni florin from Paeonia lactiflora,isochlorogenic acid C from honey suckle,ammonium gly cyrrhizinate from Glycyrrhiza uralensi s and ginsenoside Rb1 from ginseng.Results the content of paeoniflorin,isochlorogenic acid C,ammonium gly cyrrhizinate and ginsenoside Rb1 should not be less than 0.53%,0.095%,0.076%and 0.007%,the quality of Tuoli Xiaodu powder can be controlled by content determination.At the same time,the UPLC fingerprint of Tuoli Xiaodu powder was established,17 common characteristic peaks of 15 batches of Tuoli Xiaodu powder were screened out,and the medicinal materials were assigned.At the same time,the components represented by 10 characteristic peaks were identified.Through similarity evaluation,the quality differences of different batches of Tuoli Xiaodu powder were intuitively understood.Conclusion:UPLC-Q-TOF/MS technology can quickly analyze and characterize the chemical components in Tuoli Xiaodu powder.Network pharmacology predicts the targets and pathways of the main ingredients of Tuoli Xiaodu powder.The quality markers were determined by combining the two methods.To provide a basis for the research on the quality standard of Tuoli Xiaodu powder;establish the HPLC multi-index content determination of Tuoli Xiaodu powder,realize the quality control of the source medicinal materials,and establish the UPLC fingerprint of Tuoli Xiaodu powder,the difference between different batches can be visually analyzed,and the combination of the two can fully control the quality of Tuoli Xiaodu powder.
