Objective:The chemical components of Wujia biochemical capsule were determined.The target and pathway of the action were estimated by the network pharmacology,and the material basis of its efficacy was predicted.The content determination method and fingerprint analysis method of multiple index components of Wujia biochemical capsule were established,and the quality control standard of Wujia biochemical capsule was improved comprehensively.Methods:1.Chemical composition analysis of Wujia Shenghua capsule: uplc-q-tof-ms technology was used to analyze Wujia Shenghua capsule.Firstly,the preparation and analysis methods of the test sample were optimized,the experimental conditions were determined,and the data were collected.The obtained data and the self built primary mass spectrometry database were imported into the software for analysis.The molecular formula was determined by isotope abundance ratio and accurate mass number,and then compared with the retention time and secondary mass spectrometry information of the reference substance to determine the composition and infer the pyrolysis law.2.Network pharmacology research of Wujia Shenghua capsule: the obtained chemical components were screened on tcmsp platform,the relevant targets of screened components were obtained by pharmmapper server,and the related disease targets were obtained by drugbank,genecards,and digenet disease databases,and the intersection targets of the two were obtained,which were imported into Cytoscape software to construct the network diagram of "traditional Chinese medicine component target" The protein-protein(PPI)interaction network map was drawn by importing string platform,and go biological process enrichment analysis and KEGG metabolic pathway enrichment analysis were carried out by importing David platform.3.Study on quality standard of Wujia Shenghua capsule: according to network pharmacology,the component with higher degree value was selected as the active component of Wujia Shenghua capsule,and the index was selected through pre experiment Ingredients.The HPLC method was established to determine the contents of the components in 10 batches of Wujia Shenghua capsules.At the same time,the HPLC fingerprint of 10 batches of Wujia Shenghua capsules was established and introduced into the similarity evaluation system of traditional Chinese medicine chromatographic fingerprint,and the similarity was evaluated.Results:1.Chemical composition analysis of Wujia Shenghua capsule: a total of 90 compounds were identified,including 24 flavonoids,8 coumarins,9 triterpenoid saponins,9fatty acids,8 lactones,8 organic acids,6 gingerols,5 lignans,2 alkaloids,2 steroids,1glycoside and 8 other compounds.Among them,flavonoids mainly come from licorice,lignans mainly come from Acanthopanax senticosus,coumarins mainly come from Acanthopanax senticosus,Angelica sinensis and Ligusticum chuanxiong,organic acids mainly come from Ligusticum chuanxiong and Angelica sinensis,terpenoids from licorice,lactones from Ligusticum chuanxiong and Angelica sinensis,fatty acids from Ligusticum chuanxiong and peach kernel,Steroids are from Angelica sinensis and peach kernel,alkaloids are from peach kernel,Ligusticum chuanxiong and acanthopanax senticosus,glycosides are from peach kernel,gingerols are from dried ginger,and other components include all herbs.2.Network pharmacology research of Wujia Shenghua capsule: 28 components were selected from the chemical constituents of Wujia Shenghua capsule in vitro,3603 component targets were obtained through pharmmapper server,396 after weight removal,2387 disease targets were obtained through drugbank,genecards and digenet disease database,1939 after weight removal.A total of 179 overlapping targets were obtained from the intersection of component targets and disease targets.The network diagram of "medicine component target" was constructed,and 10 active components were selected according to the degree value.Then go enrichment analysis and KEGG pathway enrichment analysis were performed.3.Study on quality standard of Wujia Shenghua capsule: the pre experiment of 10 active ingredients with higher degree value selected by network pharmacology,combined with the results of pre experiment.Finally,the determination methods of six components were established.The constituents were syringin and E from Acanthopanax senticosus,ligustilide A and ligustilide from Angelica sinensis and Ligusticum chuanxiong,chlorogenic acid from Acanthopanax senticosus,Angelica sinensis and Ligusticum chuanxiong,and ammonium glycyrrhizinate from Glycyrrhiza uralensis.Then 10 different batches of Wujia biochemical capsules were determined.The content of syringin,chlorogenic acid,acanthopanax senticosus e,Ammonium Glycyrrhizinate,ligustilide A and ligustilide should not be less than 0.29%,0.25%,0.13%,0.17%,0.09%and 0.11%,respectively.The quality standard control of Wujia Shenghua capsule was achieved by the determination of the above index components.At the same time,the HPLC fingerprint of Wujia Shenghua capsules was established,and 24 common characteristic peaks of 10 batches of Wujia Shenghua capsules were selected,and their medicinal materials were assigned.At the same time,the components represented by 8 characteristic peaks were identified.Conclusion:UPLC-Q-TOF-MS can be used to analyze and characterize the chemical constituents of Wujia Shenghua capsules efficiently,simply and rapidly;It provides chemical basis for network pharmacology research;Based on the network pharmacology research method,we speculated the target and pathway of the active ingredients of Wujia Shenghua capsule,and predicted its pharmacodynamic material basis.The combination of the two provided a theoretical basis for the quality standard research of Wujia Shenghua capsule;Finally,the HPLC multi index content determination of Wujia Shenghua capsule was established to control the quality from a variety of components.At the same time,the HPLC fingerprint of Wujia Shenghua capsule was established to intuitively find the differences between different batches and realize the comprehensive control of the quality standard of Wujia Shenghua capsule.
