| Objective: More and more studies have shown that KLF15 plays a role in the protection of multiple organs in the body,but recent studies have shown that KLF15 may be related to ventricular arrhythmia(VA).This study explored the relationship between KLF15 overexpression and QT interval and VA,and provided experimental references for possible adverse reactions while KLF15 played a protective role.Methods: Thirty healthy male SD rats were randomly divided into KLF15 overexpression group(group A),empty adeno-associated virus group(group B),and saline group(group C).Each group was injected into the tail vein KLF15 overexpression of adeno-associated virus,empty adeno-associated virus and equal volume of normal saline were transfected into the myocardium to construct a KLF15 overexpression model.After 28 days,record the lead II electrocardiogram of the rat to observe the occurrence of arrhythmia in the rat and measure the electrocardiogram heart rate,QT interval and other parameters.The corrected QT interval is calculated according to the Bazett formula: QTc = QT /(RR)1/2,and the severity of the arrhythmia is evaluated according to the VA score.After the electrocardiogram is recorded,the left ventricle or the myocardial tissue from the free wall of the ventricle is taken,using Western-Blot(WB)method to detect the KLF15 protein of the three groups of rats,and RT-PCR to detect the expression of KLF15 m RNA of the three groups of rats.Establish an amputation acute traumatic stress(ATS)model,record the electrocardiogram after amputation,observe the occurrence of arrhythmia in the rat,measure the rat’s heart rate,QT interval and other parameters,calculate the QTc interval,and perform the VA score to understand the rat’s susceptibility to VA.Results: KLF15 overexpression of adeno-associated virus,empty adeno-associated virus and equal volume of normal saline were injected via tail vein to transfect myocardium to construct a KLF15 overexpression model.The results of WB and PCR indicated that the expression level of KLF15 in group A was significantly up-regulated(P<0.001),and the QT interval and QTc interval were significantly shortened(P<0.001),and there was no significant difference in VA(P>0.05);after ATS,the electrocardiogram of group A showed that the QT interval and QTc interval were significantly shortened(P<0.001),and the VA score was significantly increased(P<0.01).Conclusion: While KLF15 exerts a protective effect,it may cause QT shortening and increase VA susceptibility. |
