Aging is a degenerative change in the morphology,structure and physiological function of various cells,organs and tissues after the development and maturity of the organism.And it is a biological process affected by a variety of factors.With the aging of word’s people and the increasing incidence rate of age-related diseases,the social and economic burden brought by them are also increasing.Therefore,it is of great significance to improve the quality of life by conducting researches about the mechanism of aging,developing drugs for delaying aging and reducing the incidence rate of aging related diseases.Traditional Chinese medicine has obvious advantages in the prevention and treatment on complex diseases because of the characteristics of multi-component,multi-target and multiple ways of action.The classic famous recipe “Danggui Buxue Decoction” originated from "Differentiation of Internal Discomfort and External Injuries" was written by Li Dongyuan,a famous doctor in Jin Dynasty,which has a variety of pharmacological activities.In previous studies,it has been reported that Danggui Buxue Decoction has anti-aging activity.However,there are few researches on its anti-aging action mechanism.Therefore,in this paper,network pharmacology method was used to predict and verify the anti-aging action mechanism of Danggui Buxue Decoction from the a holistic and systematic point of view.In this study,the prediction model of anti-aging activity of Danggui Buxue Decoction was established by network pharmacology method in order to reduce the blindness of research and improve the efficiency of research.The molecular biology technology was used to verify the prediction model.Meanwhile,the effects of Danggui Buxue Decoction on liver and kidney,hematopoiesis and immune system of aging rats were studied.The anti-aging action mechanisms of Danggui Buxue Decoction were systematically reasearched by combining network pharmacology technology with traditional molecular biology technology.The network pharmacology method was applied to predict the molecular action mechanism of Danggui Buxue Decoction in delaying aging.The results showed that the main active components of Danggui Buxue Decoction were quercetin,stigmasterol,β-sitosterol,kaempferol,which mainly acted on IL6,IL10,TNF,VEGFA,AKT1,TP53,SERPINE1,PPARG,HIF1 A and other targets.Danggui Buxue Decoction could inhibit inflammation caused by aging,regulate autoimmunity and regulate cell cycle by regulating JAK-STAT signaling pathway,PI3K-AKT signaling pathway,TNF signaling pathway,TCR signaling pathway,VEGF signaling pathway,m TOR signaling pathway,etc.The antioxidant and lipid peroxidation indexes in liver,kidney and serum of aging rats were detected.The results showed that Danggui Buxue Decoction could increase the content of SOD and GSH in the liver,kidney and serum of aging rats while reduce the level of MDA.Above results suggested that Danggui Buxue Decoction played the role in delaying aging by enhancing the antioxidant capacity of the body and reducing the oxidative damage of the body.We studied the effect of Danggui Buxue Decoction on liver and kidney function of aging rats.We found that Danggui Buxue Decoction decreased the contents of ALT,AST,BUN and CRE in serum,alleviated the inflammation and fibrosis of kidney to achieved protected effect on the liver and kidney.The effect of Danggui Buxue Decoction on bone marrow hematopoietic function of aging rats was observed.The results displayed that Danggui Buxue Decoction could improve the hematopoietic function of bone marrow,increase the number of WBC,RBC,PLT in peripheral blood and BMNC cells in bone marrow,reduce the expression of CD44 and CD49 d of Sca-1+BMNC in bone marrow,and promote BMNC arrested in G1 phase to enter S phase of BMNC.Above results indicated that Danggui Buxue Decoction delayed senescence by enhancing cell proliferation and improving hematopoietic function.Danggui Buxue Decoction lessened the inflammatory injury by decreasing the release of IL-6 and TNF-α and increasing the expression of IL-10.And it affected the immune function by reducing the expression of Fas / Fas L and inhibiting the apoptosis of T lymphocytes.Moreover,it played a role in delaying aging by regulating the protein expression of PI3K-AKT/CDK-Rb pathway and PI3K-AKT/MDM2-p53 pathway. |
