Objective:By observing the cryopreservation result of different concentrated fuzi-polysaccharide on the preservation of blood vessels,the protective effect of FPS on cryopreservation blood vessels are preliminary discussed.Methods:The abdominal aortas were harvested from 72 8-week-old male SD rats.The 72 abdominal aortas were randomly divided into blank control group(Sterile Water for Injection group,10% dimethyl sulfoxide group)and experimental research group(FPS 0.1mg/m L group,FPS 1 mg/m L group,FPS 10 mg/m L group,FPS 20 mg/m L group),each group was 12 and stored directly into the liquid nitrogen after the program cools down.After one week the tissue structure of the abdominal aorta were observed by optical microscope.The expression of vascular endothelial growth factor in abdominal aorta was observed by immunohistochemistry.The apoptosis of vascular cell was testing with TUENL.Results:HE staining showed that the abdominal aortas of FPS group compared to the sterile water for Injection group appeared regular and continuously vascular endotrical cell and well structured smooth muscle fibers.Vascular VEGF detected by immunohistochemistry showed that compared with the experimental control group(sterile water for Injection group,10% dimethyl sulfoxide group)and experimental research group(FPS 0.1 mg/m L group,FPS 1 mg/m L group),the MOD of VEGF in the FPS 10 mg/m L group was significantly increased;Compared with FPS 20 mg/m L group,the MOD of VEGF increased in the FPS 10 mg/m L group.The TUNEL stained positive apoptotic cells show brown color,Hematoxylin stained cell nucleus blue.Compared with the sterile water for Injection group,the vascular apoptosis rate was significantly reduced in the 10%dimethyl sulfoxide group,FPS 0.1mg/m L group,the FPS 1mg/m L group,the FPS 10mg/m L group,the FPS 20 mg/m L group.Compared with the 10% dimethyl sulfoxide group,the vascular apoptosis rate was reduced in the FPS 10 mg/m L group and FPS 20mg/m L group.Compared with the FPS 0.1mg/m L group,the vascular apoptosis rate was significantly reduced in the 10% dimethyl sulfoxide group,FPS 1mg/m L group,the FPS10 mg/m L group,the FPS 20 mg/m L group.Compared with the FPS 1mg/m L group,the vascular apoptosis rate was significantly reduced in the FPS 10 mg/m L group,the FPS 20mg/m L group.Compared with the FPS 20 mg/m L group,the vascular apoptosis rate was reduced in the FPS 10 mg/m L group.Conclusion:The fuzi-polysaccharides have a protective effect on the cryopreservation of the abdominal aorta.The high concentration group of fuzi-polysaccharides had better effect on the cryopreservation of the abdominal aorta than the low concentration of fuzi-polysaccharides.The fuzi-polysaccharides concentration of 10 mg/m L may be better preserved in the cryopreservation of the abdominal aorta.
