Proteins and polysaccharides are the main bearers of body composition and biological functions.Human serum albumin(HSA)is the most abundant protein in plasma,which plays a key role in transporting drugs and maintaining blood osmotic pressure.The level of HSA is closely related to the health of the body.For example,the level of HSA in blood and urine is an important reference index for cardiovascular and renal diseases and an important parameter for preclinical diagnosis.For the quantitative analysis of HSA,using the conjugated skeleton of pyrene and cyanobenzene as the light-emitting unit,a new type of negatively charged fluorescent probe PYPOS was designed and synthesized to examine its photophysical properties and its effect on common proteins(such as human serum).The quantitative performance of albumin,insulin,esterase,trypsin,lysozyme,etc.)established a rapid quantitative method for HSA in urine.PYPOS has typical aggregation-induced luminescence(AIE)properties,exhibits highly sensitive,fast and stable fluorescence response to various protein molecules,and is not interfered by ions,polysaccharides,vitamins and amino acids during the detection process.After PYPOS is combined with protein molecules,the color and fluorescence show significant changes,which can be used for protein colorimetric and fluorescence quantitative analysis.Experimental results show that PYPOS can be used for accurate quantification of HSA in urine,and can also be used for the identification of clinical samples of different urine protein grades,providing a convenient and effective tool for rapid detection of proteinuria and home monitoring of patients with kidney disease.Heparin(Hep) is the glycosaminoglycan with the highest charge density in the body.It plays an important role in the blood anticoagulation process.It is the most widely used drug for preventing coagulation during surgery and treating thrombotic diseases.However,overdose of Hep will cause side effects such as bleeding,thrombocytopenia and hyperkalemia.Therefore,the development of efficient and convenient analysis and detection methods for Hep is of great significance for disease prevention and early diagnosis.For the detection of Hep in serum,a new type of fluorescent probe PYPN with two positive charges was designed and synthesized with pyrene and pyridine as the basic building blocks.The photophysical properties and the analytical performance of Hep were investigated.Fluorescence analysis method of heparin in serum.PYPN has the following characteristics in Hep detection: fluorescence emission at 500-700 nm,can effectively reduce the interference of serum matrix background fluorescence;response time within 30 seconds,suitable for rapid detection on site;color and fluorescence changes obviously,suitable for naked eye detection;Good selectivity,less interference by ions,thiols,albumin,ATP and other glycosaminoglycans.PYPN can be used to accurately quantify heparin in the serum of humans,rabbits,cattle and other animals,with a detection limit of 0.74-0.82 μg/m L. |
