Objective: To study the reproductive toxicity and the mechanism of Successive generations of exposure to dibutyl phthalate in model organism Caenorhabditis elegans.Methods: We set up blank control group(blank group),vehicle group(0.1% DMSO)and experimental group(0.1mg/L,1mg/L,10mg/L and 100mg/L DBP groups).The control group,0.1% DMSO group,0.1mg/L,1mg/L,10mg/L and 100mg/L DBP groups were exposed for 5 generations.One L1 stage elegans was taken from each concentration group and repeated for 15 times in each group.The instantaneous oviposition rate,number of offspring and generation time were determined in each generation.One L1 stage elegans was exposed to 0.1mg/L DBP for 10 generations.After about 2000 elegans of the 10 th generation propagated,RNA was extracted and analyzed.Results: 1.The results in the five generations of exposure experiments showed that the instantaneous oviposition rate on the first day in 1mg/L and 100mg/L DBP groups was significantly decreased compared with the blank group in the 1st generation(P<0.05).Compared with the blank group,the instantaneous oviposition rate on the first day in the0.1mg/L,1mg/L,10mg/L and 100mg/L DBP groups was significantly lower(P<0.01)in the 4th generation.Compared with the 1st generation,the instantaneous oviposition rate on the second day of the 3rd,4th and 5th generation decreased significantly at 0.1mg/L,1mg/L,10mg/L and 100mg/L DBP group(P<0.01).Compared with the blank group and0.1%DMSO group,the instantaneous oviposition rate on the second day decreased significantly in 0.1mg/L,1mg/L,10mg/L and 100mg/L DBP groups at tht 2nd,3rd,4th,5th generation(P<0.05).Compared with the blank group,the number of offspring in 0.1mg/L,1mg/L,10mg/L and 100mg/L DBP group significantly reduced the number of offspring in2 nd,3rd,4th and 5th generation(P<0.01).Compared with the 1st generation,the generation time of the 4th and 5h generation was significantly longer in the generation time at the0.1mg/L,1mg/L,10mg/L and 100mg/L DBP group(P<0.05).In the 1st,2nd,3rd,4th and5 th generation,the generation time of 0.1mg/L,1mg/L,10mg/L and 100mg/L DBP group was significantly longer than that of the blank group(P<0.01).2.The effect of continuous10 generations exposure to blank group and 0.1mg/L DBP group showed that the number of differential genes increased was 862,and the number of differential genes decreased was 1208.The expression levels of apoptosis in elegans tissues were detected,which showed that the expression of nuc-1 and asns-1 decreased significantly.The expression levels of oxidative stress in elegans tissues were detected,which showed that the expression levels of sod-5 and hsp-16.1 increased significantly and the expression levels of sod-1 and klo-2 decreased significantly.Conclusion: successive generations of exposure to dibutyl phthalate decrease the reproductive capacity of elegans,raise the expression of spo-11,lgg-1,sod-5,hsp-16.1,reduce the expression of lin-3,haf-4,acs-1,let-767,nuc-1,asns-1,sod-1,klo-2,and may be closely related to the apoptosis and oxidative stress of elegans.
