| In recent years,despite the continuous development of life science and medical science,there are still many diseases that threaten human life and health.Among the most common malignant tumors among women in the world,cervical cancer(CC)is the second most common cancer.Although screening for cervical cancer has been gradually carried out,the incidence and mortality of cervical cancer are still at a high level in some developing countries.Therefore,it is necessary to deeply understand the role of cervical cancer on the occurrence and development of the disease from the level of gene expression and methylation of cervical cancer to analyze its impact on the survival of cervical cancer patients.Firstly,the gene expression,DNA methylation and clinical data of cervical cancer were downloaded from the TCGA database,and univariate Cox regression analysis was performed based on the differentially expressed genes of cervical cancer.The obtained genes were analyzed by GO and KEGG,and 11 differentially expressed genes were found to be closely related to DNA replication and repair.Then Kaplan-Meier survival analysis was performed on the patients using these 11 genes,and 6 high expression genes,MCM2,MCM5,PCNA,POLA2,RFC5 and RNASEH2 A,may have the potential effect on the survival rate of cervical cancer patients.Secondly,the WGCNA analysis based on the differentially expressed genes and clinical characteristics of cervical cancer were performed after the patients grouped by age,and found that,The genes in the black module in youth group are related to the genetic vascular system and the proliferation and migration of endothelial cells.The genes in the red module in middle-aged group and the turquoise module in old-aged group are related to epidermal cells and keratinization,which indicates that smoking has a greater impact on patients of middle-aged group and old-aged group.Finally,the prognosis model was build using the LASSO and COX regression algorithm and difference of gene methylation,based on the analysis of Kaplan-Meier in patients with cervical cancer.It was found that the survival rate of high-risk group is much lower than that of low-risk group,the performance evaluation model of receiver-operating characteristic curve(ROC)was used,and the area under the curve(AUC)was 0.73,which has a certain predictive value.Patients were grouped according to age and evaluated by Kaplan-Meier and ROC curves.It was found that the predictive power of the model in middle-aged group was general,but more significant in other age groups.Then,the correlation between methylated genes in the model and their expression data was analyzed to explore the correlation strength between gene methylation and expression and its influence on the occurrence and development of cervical cancer.These conclusions may have potential significance for the clinical diagnosis of cervical cancer. |
