Objective: To investigate the expression and significance of Connexin 43 and matrix metalloproteinase 1 in the fetal membranes of women with premature rupture of fetal membrane in Xining area.Methods: Collect the tissue specimens of premature membranes of women who gave birth in the hospital of Qighai University from December 2019 to May 2020(PROM group)and healthy women who gave birth naturally(control group)during the same period.Fetal membrane tissue specimens were taken within 15 minutes of delivery of the placenta from the placenta attached side 2cm away from the fetal membrane rupture.There were 30 cases in the PROM group and 30 cases in the control group.Western blot experiments and immunohistochemical staining experiments were performed on fetal membrane tissue samples to detect the intracellular localization and protein expression levels of CX-43 and MMP-1 in fetal membrane samples.Data analysis adopts t test,and correlation analysis adopts Person correlation test.Results: By studying the location and expression of CX-43 and MMP-1 in the fetal membrane tissues of PROM and healthy women in Xining area,the following results were obtained.(1)Through immunohistochemical staining experiments,the results showed that CX-43 is mainly located in the cell membrane of the chorionic layer,and MMP-1 is mainly diffused in the cytoplasm and part of the nucleus of the chorionic cell layer and amniotic layer;in the fetal membrane of the PROM group Compared with the control group,the AOD value of CX-43 was significantly different(P<0.05);the AOD value of MMP-1 in the fetal membrane of the PROM group was compared with the control group,and the difference was not statistically significant(P>0.05).(2)Western blot experiment showed that the protein expression level of CX-43 in the fetal membrane of the PROM group was compared with that of the control group,the difference was statistically significant(P<0.05);the protein expression level of MMP-1 in the PROM group was compared with the control group In comparison,the difference was not statistically significant(P>0.05).(3)Pearson correlation analysis between CX-43 and MMP-1 protein expression was performed in PROM group and control group.Both CX-43 and MMP-1 protein expression were negatively correlated in the two groups.Theoretical and practical significance: By studying the location and expression of CX-43 and MMP-1 in the fetal membrane tissues of PROM and healthy women in Xining area,the study found that:(1)CX-43 and MMP-1 are both expressed in the fetal membrane tissues.CX-43 is mainly located in the cell membrane of the chorionic layer,and MMP-1 is mainly diffused in the cytoplasm and part of the nucleus of the chorionic cell layer and the amniotic layer.(2)The protein expression of CX-43 in the fetal membrane tissue of PROM women was statistically significant compared with that of control women,while the protein expression of MMP-1 in PROM women’s fetal membrane tissue was not statistically significant compared with control women.It shows that the occurrence of premature rupture of membranes in Xining area may be related to the decreased expression of CX-43,and CX-43 may become an indicator for the prevention and treatment of PROM in Xining area.(3)Pearson correlation analysis found that CX-43 is negatively correlated with MMP-1protein expression,or it can be inferred that in the occurrence of premature rupture of membranes in Xining area,the decrease of CX-43 level in membrane tissue is related to the level of MMP-1,but Due to the small sample size in this experiment,the correlation between MMP-1 and CX-43 in patients with premature rupture of membranes in Xining area needs to be further studied.
